茯苓多糖对非小细胞肺癌NCI-H460细胞的抑制作用

为评价获苓(Poria cocos)多糖对非小细胞肺癌NCI-H460细胞的抑制作用,用CCK-8法测定茯苓多糖对细胞增殖的抑制率,用DAPI染色法、AOEB双荧光染色法分析茯苓多糖对细胞凋亡的影响,用细胞划伤愈合实验测定茯苓多糖对细胞迁移能力的影响,用蛋白免疫印迹实验检测茯苓多糖对细胞凋亡、迁移相关蛋白表达的影响.结果表明:茯苓多糖具有抑制NCI-H460细胞增殖、集落形成和迁移的能力,可诱导NCI-H460细胞凋亡;500、1000 μg·mL-1的茯苓多糖可使Bax和P53蛋白表达量升高,Bcl-2和MMP-2蛋白表达量降低.
Inhibitory Effect of Poria cocos Polysaccharide on Non-Small-Cell Lung Cancer Cell Line NCI-H460
The inhibitory effect of Poria cocos polysaccharide(PCP)polysaccharide on non-small-cell lung cancer NCI-H460 cells was studied.CCK-8 kit was used to determine the inhibition rate of PCP on NCI-H460 cell proliferation.DAPI staining and AOEB double fluorescence staining were used to analyze the effect of PCP on apoptosis in NCI-H460 cells.The scratch assay was used to evaluate the effect of PCP on cell migration.Western blotting was used to detect the effect of PCP on protein expression pertaining to apoptosis and migration.The results showed that PCP effectively inhibited the proliferation,colony formation and migration of NCI-H460 cells,and induced cell apoptosis.At 500 and 1 000 μg·mL-1,PCP significantly increased the expression of Bax and P53,but inhibited the expression of Bcl-2 and MMP-2 in NCI-H460 cells.
