Inhibitory Effect of Poria cocos Polysaccharide on Non-Small-Cell Lung Cancer Cell Line NCI-H460
The inhibitory effect of Poria cocos polysaccharide(PCP)polysaccharide on non-small-cell lung cancer NCI-H460 cells was studied.CCK-8 kit was used to determine the inhibition rate of PCP on NCI-H460 cell proliferation.DAPI staining and AOEB double fluorescence staining were used to analyze the effect of PCP on apoptosis in NCI-H460 cells. The scratch assay was used to evaluate the effect of PCP on cell migration.Western blotting was used to detect the effect of PCP on protein expression pertaining to apoptosis and migration. The results showed that PCP effectively inhibited the proliferation, colony formation and migration of NCI-H460 cells, and induced cell apoptosis. At 500 and 1 000 μg·mL-1,PCP significantly increased the expression of Bax and P53,but inhibited the expression of Bcl-2 and MMP-2 in NCI-H460 cells.
