Immunoregulatory Activity of Hypsizygus ulmarius Fruiting Body and Its Extracts
In order to study the immunomodulatory effects of Hypsizygus ulmarius fruiting body powder and its different extracts on mice,eight experimental groups were set up as follows:high-dose(1.2 g·kg-1)and low-dose(0.3 g·kg-1)fruiting body powder groups,high-dose(1.392 g·kg-1)and low-dose(0.348 g·kg-1)petroleum ether extract groups,high-dose(3.306 g·kg-1)and low-dose(0.826 g·kg-1)ethyl acetate extract groups,and high-dose(28.78 g·kg-1)and low-dose(7.19 g·kg-1)water extract groups.These powders and extracts were administered to mice with spleen deficiency syndrome by gavage,respectively.Then mice in different treatment groups were measured for body mass,spleen index,and serum contents of interleukin-2(IL-2),interleukin-6(IL-6),immunoglobulin M(IgM),interferon-y(IFN-γ),D-xylose,albumin(ALB)and pancrelipase(PL).Hematoxylin-eosin staining was used to observe the microscopic structure of mouse spleen tissue.RT-qPCR was used to detect the gene expression of vitamin D-binding protein(DBP)and occludin(OCLN),and Western blotting was used to detect and semiquantify DBP and OCLN.The results showed that the body mass of mice in all treatment groups significantly increased compared with the model group at 14 d.The spleen index of the high-dose fruiting body powder group was significantly higher than that of the model group.All treatment groups showed higher contents of D-xylose,IFN-γ,ALB,IL-2,IgM and PL than the model group.In contrast,IL-6 level was significantly decreased in all treatment groups compared with the model group.Compared with the model group,the relative expression of DBP gene was significantly increased in all high-dose groups,but significantly decreased in all low-dose groups.The relative expression of OCLN gene was significantly increased in the low-dose powder group,petroleum ether extract groups,and water extract groups.The protein levels of DBP and OCLN were significantly increased in all treatment groups.These results suggested that H.ulmarius fruiting body powder and its extracts have immunomodulatory and spleen-invigorating effects,and thus provided a reference for further development and utilization of the mushroom.
