Identification and Expression Pattern Analysis of BAG Family in Pleurotus ostreatus
The BAG domain-containing proteins(BAGs)of Pleurotus ostreatus(PoBAGs)were retrieved from NCBI and JGI databases,analyzed by bioinformatics tools,and then measured for expression at different developmental stages(mycelium,primordium,fruiting body)and under temperature stresses(high temperature at 42 ℃and low temperature at 4 ℃).The results showed that a total of three proteins,named PoBAG-1,PoBAG-2 and PoBAG-3,contain the BAG domain.In addition,PoBAG-1 and PoBAG-2 also contain an IQ motif and a ubiquitin-like(UBL)domain,respectively.PoBAG-1 and PoBAG-3 are hydrophilic unstableand phylogenetically close.They were predicted to be mainly localized in the nucleus and cytoplasm.PoBAG-2 is a hydrophilic stable protein closely related to BAG proteins with UBL domain in other organisms.It was predicted to be mainly localized in mitochondria and cytoplasm.The relative expression of the three PoBAGs were significantly up-regulated in primordium and fruiting body compared with mycelium.Compared with the control(25 ℃),high temperature(42 ℃)stress resulted in extremely significantly increased relative expression levels of PoBAG-1 and PoBAG-3,and significantly decreased PoBAG-2 expression.Under low temperature(4 ℃)stress,the relative expression levels of PoBAG-1 and PoBAG-2 were significantly increased while PoBAG-3 was significantly down-regulated.This study provided a reference for further analysis of the function of PoBAG family proteins in the growth and development of P.ostreatus,and its response to temperature stress.
