摘要

测定中国块菌(Tuber sinense)营养成分,将小鼠随机分为对照组(基础饲料)、实验Ⅰ组(99%基础饲料+1%中国块菌粉末)、实验Ⅱ组(95%基础饲料+5%中国块菌粉末),测定12、24、36 d小鼠脾脏、胸腺指数,白细胞数量,五类白细胞、T淋巴细胞百分比以及血清中IgG含量,并检测在36 d时小鼠脾脏中肿瘤坏死因子α(TNF α)、白介素1β(IL-1β)、白介素6(IL-6)基因的表达量.结果表明:与对照组比较,实验Ⅱ组的脾脏指数在36 d时显著降低;实验Ⅰ组的白细胞总数在12、24、36 d时显著增加;实验Ⅱ组的单核细胞百分比在24、36 d时显著增加;实验Ⅰ组的嗜中性粒细胞、淋巴细胞百分比在24、36 d时显著增加,实验Ⅱ组的在12、24、36 d时显著增加;实验Ⅱ组的T淋巴细胞百分比在24、36 d时显著增加;实验Ⅰ组的血清中IgG含量在36 d时显著增加,实验Ⅱ组的在12、24、36d时显著增加;实验Ⅰ组、实验Ⅱ组的脾脏中TNF-α表达量在36 d时显著增加.研究结果可为中国块菌的开发利用提供参考.

Abstract

The nutrient composition of Tuber sinense was determined,and then its fruiting body powder was added to the basic mouse feed at 1%(test group Ⅰ,1%T.sinense+99%basic diet)and 5%(test group Ⅱ,5%T.sinense+95%basic diet),respectively.Mice fed with the basic feed served as the normal control group.At 12 d,24 d and 36 d,mice in different groups were measured for spleen index,thymus index,leukocyte count,percentages of five types of leukocytes,the percentage of T lymphocytes,and serum IgG level.At 36 d,relative expression levels of TNF-α,IL-1βand IL-6 in spleen were also determined.The results showed that compared with the control group,the spleen index of group Ⅱ was significantly decreased at 36 d;the number of leukocytes in group Ⅰ was significantly increased at 12 d,24 d and 36 d;the percentage of monocytes in group Ⅱ was significantly increased at 24 d and 36 d;both group Ⅰ andⅡ had significantly increases in percentages of neutrophils and lymphocytes(24 d and 36 d for group Ⅰ,and 12 d,24 d and 36 d for group Ⅱ);the percentage of T lymphocytes in group Ⅱ was significantly increased at 24 d and 36 d;the serum IgG content in group Ⅰ was significantly increased at 36 d,and that of group Ⅱ was significantly increased at 12 d,24 d and 36 d;the relative TNF-α expression in spleen was significantly increased in both group Ⅰ and Ⅱ at 36 d.The results provided a reference for the development and utilization of T.sinense.
