术前应用甲壳素衍生物对瘢痕疙瘩组织中TSG6-P38/JNK信号通路的影响

摘要
目的 探究术前应用甲壳素衍生物对瘢痕疙瘩中肿瘤坏死因子α刺激基因 6(TSG-6)-p38/c-Jun氨基末端激酶(JNK)通路的影响.方法 选择自 2019 年 10 月至 2021 年 8 月,河北北方学院附属第一医院皮肤科收治的 79 例瘢痕疙瘩患者作为研究对象,接受手术切除治疗并根据术前是否使用甲壳素衍生物分为甲壳素组(42 例)和对照组(37 例),甲壳素组术前给予甲壳素凝胶涂抹,2 次/d,连续 14 d;对照组术前不接受任何治疗.手术中收集瘢痕疙瘩组织,采用HE染色检测病理改变,Western blot检测PCNA、Ki-67、TSG6、p-p38、p-JNK、B淋巴细胞瘤-2 基因(Bcl-2)、Bcl-2 相关X蛋白(Bax)、裂解型caspase-3(Cleaved caspase-3)的表达水平,采用TUNEL法检测细胞凋亡率.结果 与对照组比较,甲壳素组瘢痕疙瘩中炎症细胞浸润减少,PNCA、Ki-67、Bcl-2 的表达水平降低,细胞凋亡率及TSG-6、Bax、Cleaved caspase-3 的表达水平增加(P<0.05).结论 甲壳素衍生物调控瘢痕疙瘩中的细胞增殖及凋亡,这一作用与调控TSG6-p38/JNK通路有关.
Abstract
Objective To study the regulation effect of preoperative application of chitin derivatives on Tumor necrosis factor-al-pha stimulated gene-6(TSG-6)-p38/c-Jun N-terminal kinase(c-Jun N-terminal kinase,JNK)pathway in keloid.Methods Keloid pa-tients treated in our hospital from October 2019 to August 2021 were selected as the research object.They underwent surgical resection and were divided into the chitin group(n=42)and control group(n=37)according to whether treated with chitin derivatives.The chitin group was given chitin gel twice a day for 14 consecutive days before the operation,while the control group did not receive any treatment before the operation.During surgery,the keloid tissues were collected.The pathological changes were detected by HE staining,the expression levels of PCNA,Ki-67,and the apoptosis rate were detected by the TUNEL method.Results Compared with the control group,the infil-tration of inflammatory cells decreased,the expression level of PCNA,Ki-67,Bcl-2 decreased,and the apoptosis rate and the expression levels of TSG-6,Bax and cleaved caspase-3 increased in keloid of chitin group(P<0.05).Conclusion Chitin derivatives regulate cell proliferation and apoptosis in keloid,which is related to the regulation of the TSG6-p38/JNK pathway.
