Effect of preoperative application of chitin derivatives on TSG6-p38/JNK pathway in keloid
Objective To study the regulation effect of preoperative application of chitin derivatives on Tumor necrosis factor-al-pha stimulated gene-6(TSG-6)-p38/c-Jun N-terminal kinase(c-Jun N-terminal kinase,JNK)pathway in keloid. Methods Keloid pa-tients treated in our hospital from October 2019 to August 2021 were selected as the research object. They underwent surgical resection and were divided into the chitin group(n=42)and control group(n=37)according to whether treated with chitin derivatives. The chitin group was given chitin gel twice a day for 14 consecutive days before the operation, while the control group did not receive any treatment before the operation.During surgery, the keloid tissues were collected. The pathological changes were detected by HE staining, the expression levels of PCNA,Ki-67,and the apoptosis rate were detected by the TUNEL method. Results Compared with the control group, the infil-tration of inflammatory cells decreased, the expression level of PCNA,Ki-67,Bcl-2 decreased, and the apoptosis rate and the expression levels of TSG-6,Bax and cleaved caspase-3 increased in keloid of chitin group(P<0.05).Conclusion Chitin derivatives regulate cell proliferation and apoptosis in keloid, which is related to the regulation of the TSG6-p38/JNK pathway.
