miR-194 inhibits the formation of rabbit ear scars by inhibiting TGF-β1 expression and downregulating inflammatory factors
Objective To explore the therapeutic effect of miR-194 on hypertrophic scars in rabbit ears.Methods From Au-gust 2022 to August 2023,the Plastic Surgery and Cosmetic Center of the First Affiliated Hospital of Harbin Medical University established a rabbit ear scar model on 6 New Zealand white rabbits,and treated the rabbit ear scars by injecting miR-194.Detect the effect of miR-194 on the morphology of rabbit ear scars through gross observation,HE staining,and MASSON staining.Protein immunoblotting as-say was used to detect the effects of miR-194 on the expression of TGF-β1,VEGF,and COL-1.ELISA detection of the effect of miR-194 on the expression of TGF-β1,IL-1β and TNF-α.Results General observation showed that the scars in the treatment group are smaller,softer,and smoother,with a color closer to normal skin color.HE staining sections showed that miR-194 treatment can reduce the formation of rabbit ear scars,flatten the arrangement of fibroblasts,downregulate the release of inflammatory factors,and promote angiogenesis.The results of protein immunoblotting experiments showed that miR-194 can significantly inhibit the expression of TGF-β1、VEGF and COL-1.The ELISA results showed that miR-194 could reduce the expression of TGF-β1、IL-1β and TNF-α(P=0.02,P=0.01,P=0.02).Conclusion MiR-194 can inhibit the formation of hypertrophic scars by reducing the expression of TGF-β1,VEGF,and COL-1 in tis-sues and downregulating the activity of inflammatory factors.
