摘要
目的 利用在线数据库和生物信息学的方法,分析ZMYND8在皮肤黑色素瘤(malignant melanoma,MM)中的表达水平及其与MM发生发展、预后及免疫细胞浸润的相关性.方法 自2024年2-4月,联勤保障部队第九四○医院烧伤整形科采用GEPIA在线数据库(TCGA数据)分析多种正常组织与MM中ZMYND8表达情况,进一步利用GEO数据验证ZMYND8在MM中的表达差异;UALCAN数据库分析ZMYND8与MM患者预后的相关性;采用The Human Protein Atlas数据库分析ZMYND8在MM组织中的免疫组化表达;String数据库构建ZMYND8共表达基因的蛋白互作网络;采用TMER数据库分析ZMYND8表达与免疫浸润细胞相关性.结果 ZMYND8表达水平在MM组织中显著高于正常组织(P<0.01);ZMYND8的高转录水平与性别、年龄、肥胖情况、淋巴转移和癌症分期阶段密切相关(P<0.05);ZMYND8 mRNA表达水平与MM预后和免疫细胞浸润相关(P<0.05).结论 ZMYND8在MM中高表达,与MM的发生发展、预后及免疫细胞浸润相关,可作为皮肤MM诊断、预后及药物治疗的潜在标志物.
Abstract
Objective To use online databases and bioinformatics methods to analyze the expression level of ZMYND8 in malig-nant melanoma(MM)and its association with MM development,prognosis,and immune cell infiltration.Methods The GEPIA online database(TCGA data)was used to analyze the expression of ZMYND8 in a variety of normal tissues compared with the cancer tissues,and the GEO data was further used to confirm the distinctions in the expression of ZMYND8 in MM;the UALCAN database analyzed the corre-lation between ZMYND8 and the prognosis of MM patients;and The Human Protein Atlas database was used to to analyze the immunohis-tochemical expression of ZMYND8 in MM tissues;String database to establish the protein interactions network of ZMYND8 co-expressed genes;and TMER database to analyze the association between ZMYND8 expression and immune cells infiltrating.Results In MM tissues,the expression level of ZMYND8 was higher than that in normal tissues(P<0.01);the high transcription level of ZMYND8 was highly cor-related with gender,age,obesity status,lymphatic metastasis,and cancer staging stage(P<0.05);and the expression level of ZMYND8 mRNA was correlated with the prognosis of MM and immune cell infiltration(P<0.05).Conclusion ZMYND8 is highly expressed in MM,and its correlation with MM development,prognosis,and immune cells infiltration may serve as a potential marker for the diagnosis,prognosis,and drug treatment of MM.
