二甲双胍对裸鼠皮肤急性光损伤的保护作用研究

目的 探讨二甲双胍(metformin,MET)对紫外线所致裸鼠皮肤急性光损伤的保护作用.方法 自2022年1月至2023年1月,西南医科大学将裸鼠随机分为空白组、MET组、对照组,空白组不做任何处置,对照组仅接受65 J/cm2中长波紫外线照射,MET组接受65 J/cm2中长波紫外线照射同时给予腹腔注射MET溶液.通过大体观察对比裸鼠皮肤外观变化,通过HE染色、Masson染色对比裸鼠皮肤病理变化,通过流式ROS检测对比皮肤组织ROS变化,通过免疫印迹实验(Western blot,WB)检测炎症因子IL-1 β、IL-6、TNF-α及MMP-1表达情况.采用GraphPad Prism 8.3进行数据分析,P<0.05为差异有统计学意义.结果 MET组裸鼠背部皮肤出现的皮屑,皮肤光泽度降低、局部红肿、破溃、痂壳形成等病变较对照组轻;HE染色提示,与空白组相比,MET组和对照组镜下均出现不同程度的表皮层增厚,角质层角化过度伴角化不全,皮肤附属器减少,皮下毛细血管浸润扩张,但MET组上述表现更轻,镜下表皮厚度测定提示,MET组表皮厚度明显低于对照组(P<0.05);Masson染色提示,与空白组相比,MET组和对照组蓝染区域均有不同程度的缩小,分布欠均匀,蓝染的胶原蛋白排布杂乱,连续性被打断,但MET组上述表现更轻;流式ROS检测提示,与空白组相比MET组和对照组ROS均明显升高(P<0.05,P<0.001),与MET组相比对照组 ROS 明显升高(P<0.05);WB 提示,与空白组相比 MET 组和对照组 IL-1β(P<0.01,P<0.0001)、IL-6(P<0.05,P<0.0001)、TNF-α(P<0.05,P<0.0001)、MMP-1(P<0.05,P<0.0001)相对表达均较高;与对照组相比,MET 组 IL-1 β(P<0.05)、IL-6(P<0.05)、TNF-α(P<0.05)、MMP-1(P<0.05)相对表达较低.结论 二甲双胍对紫外线所致裸鼠皮肤急性光损伤产生了一定的保护作用.
Protective effect of metformin on acute photodamage in nude mice
Objective To explore whether metformin has protective effect on acute photodamage of nude mice induced by ultra-violet.Methods The nude mice were randomly divided into blank group,MET group and control group.The blank group does not do any processing,the MET group and control group received 65 J/cm2 wave ultraviolet irradiation,at the same time the MET group was given in-traperitoneal injection of MET solution.Then,the skin appearance changes of nude mice were compared by gross observation,the skin pathological changes of nude mice were compared by HE staining and Masson staining,the ROS of skin tissue were analyzed and compared by flow cytometry,and the expressions of IL-1(3,IL-6,TNF-α and MMP-1 were detected by Western blot.Data analysis and mapping were performed using GraphPad Prism 8.3.Results After ultraviolet irradiation at 65 J/cm2 and injection,the back skin of nude mice in the MET group showed less lesions,such as dandruff,skin glossiness,local redness,ruptures,and crust-forming.H&E staining showed that compared with the blank group,the MET group and control group showed varying degrees of epidermal thickening,hyperkeratosis with parakeratosis of the cuticle,reduction of skin appendages,and infiltration and expansion of subcutaneous capillaries.The above perfor-mance was less severe in the MET group than that in the control group.The epidermal thickness in the MET group was significantly lower than that in the control group(P<0.05).Masson staining showed that compared with the blank group,the blue-stained areas in the MET group and the control group were narrowed to varying degrees,the distribution was uneven,the blue-stained collagen was arranged disor-derly,and the continuity was interrupted.The above performance was also less severe in the MET group than in the control group.Flow cy-tometry showed that ROS in both MET and control groups was significantly higher than that in blank group(P<0.05,P<0.001),but the control group increased more(P<0.01).Western blot showed that the relative expressions of IL-1(3(P<0.01,P<0.0001),IL-6(P<0.05,P<0.0001),TNF-α(P<0.05.P<0.0001)and MMP-1(P<0.05,P<0.0001)in the MET group and control group were higher than those in the blank group.Compared with control group,the expressions of IL-1(3(P<0.05),IL-6(P<0.05),TNF-α(P<0.05)and MMP-1(P<0.05)in MET group were lower.Conclusion Metformin has certain protective effect on acute photodamage in nude mice.
