摘要
目的 改进并建立用于测定动物源性食品中维生素B2 含量的分子荧光光度法.方法 以五花肉、鸡肉、猪肝、牛肾为研究对象,参考GB 5009.85-2016《食品安全国家标准食品中维生素B2 的测定》,进一步简化实验过程、优化实验条件,分析检测过程中影响结果的多种因素,如pH、光照强度、连二亚硫酸钠对荧光猝灭的作用,提高动物源性食品中维生素B2 测定方法的准确性.结果 确定了pH 6.5时荧光强度最大,随着光照强度增强及光照时间加长荧光强度逐渐降低,5 h后维生素B2 基本分解完全.维生素B2 在0~10 μg浓度范围内线性良好,r2=0.9993,检出限为0.004 mg/100 g,定量限为0.02 mg/100 g;加标回收率范围在90%~100%.平行样品测定结果的相对标准偏差小于6%.结论 该方法准确、设备简单、成本低廉,可以满足基层实验室对动物源性食品中维生素B2 的测定需求.
Abstract
Objective To improve and establish a molecular fluorescence spectrophotometry method for determining the content of vitamin B2 in animal derived foods.Methods Using pork belly,chicken,pig liver,and cow kidney as research objects,and referring to GB 5009.85-2016 National Standard of Food Safety—Determination of Vitamin B2 in Foods,further simplified the experimental process,optimize experimental conditions,analyzed various factors that affect the results during the detection process,such as pH,light intensity,and the effect of sodium hydrosulfite on fluorescence quenching,improved the accuracy of the method for determining vitamin B2 in animal derived foods.Results The maximum fluorescence intensity was determined at pH 6.5.As the light intensity increased and the light duration increased,the fluorescence intensity gradually decreased.After 5 hours,vitamin B2 basically decomposed completely.Vitamin B2 exhibited good linearity within the range of 0~10 μg,with r2=0.9993;the limit of detection was 0.004 mg/100 g,and the limit of quantification was 0.02 mg/100 g.The recovery rates for spiking ranged from 90%~100%.The relative standard deviation of parallel sample determination results was less than 6%.Conclusion This method is accurate,simple in equipment,and cost-effective,and can meet the needs of grassroots laboratories for the determination of vitamin B2 in animal derived foods.
