Experiment Design for Colorimetric Determination of GSH Based on Surface Vulcanization Prussian Blue
In this experiment,the application of Prussian blue in biological analysis is studied.Based on the UV-Vis spectrophotometry method,combining the knowledge of chemistry and biochemistry,a surface vulcanization Prussian blue catalyst is innovatively prepared by sulfide method.The catalyst can effectively eliminate the interference of its own blue color and significantly improve the activity of peroxidase.In the presence of H2 O2,the catalyst catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine(TMB)to form blue oxidized TMB(oxTMB).Kinetic analysis shows that the catalysts have strong affinity with substrates and present excellent peroxidase activity.The surface vulcanization Prussian blue exhibits high sensitivity and selectivity in the detection of glutathione by colorimetry.This experiment including material preparation,instrument characterization and detection,can be divided into multiple modules to carry out experimental teaching in inorganic chemistry,instrumental analysis and biochemistry.It is conducive to enhancing innovative and practical ability of students,and improving the ability of integrating knowledge.
