Sample Preparation Method of Sperm for Scanning Electron Microscopy
Conventional sample preparation methods of scanning electron microscopy are prone to structural representation defects,such as sperm agglomeration and deformation.Most of the oligospermia can not meet the requirements of sample preparation,and technical improvement is urgently needed.A polyol biocompatible polymer is introduced into sperm sample preparation.The fixed sample is resuspended,evenly coated on the silicon,and directly used for scanning electron microscope observation after natural drying.The imaging quality of sperm scanning electron microscopy is dramatically improved by sample preparation exploring and optimizing.
