针对空间学习记忆测试仪器的局限性,自主研发了 1 款多功能闭合式迷宫,该迷宫由圆形实验箱体、摄像头及电路控制系统组成,可实现远程实验动物行为学监测.利用该系统对吗啡成瘾与戒断空间学习记忆行为学进行测试和验证:皮下注射吗啡10 mg/kg,每天2 次,连续9d建立吗啡成瘾大鼠模型;第12d开始自然戒断,对照组给予等体积生理盐水,比较第11、13、15、17 d 2 组动物空间认知能力变化.测试结果表明,在第11d和第 13d的观察期间,吗啡组相较于对照组在目标区域的停留时间显著增长;在第15d和第17d的观察期间,2 组实验动物在目标区域的停留时间无统计学差异,基于大鼠的成瘾和戒断行为可在系统中稳定训练完成.实验表明该多功能闭合式迷宫能满足对实验动物的空间导航认知能力的评价,且有较好的应用效果.
Evaluation and Application of Spatial Cognitive Ability in the Multi-function Closed Maze
In view of the limitations of spatial learning and memory test instruments,a multi-function closed maze is designed to realize remote experimental animal behavioral monitoring,which is composed of circular experimental box,camera and circuit control system.The system is used to test and validate the behavior of morphine addiction and spatial learning and memory with withdrawal.After subcutaneous injection of morphine 10 mg/kg twice a day for 9 days,natural withdrawal began on the day 12 and the control group receives equal volume of saline injection to compare the spatial cognitive changes on days of 11,13,15 and 17.The results show significant differences in the morphine group on days of 11 and 13 and not in the target area on days of 15 and 17.The addiction and withdrawal behavior in rats can be completed in the system.The experiment shows that the multi-function closed maze can satisfy the evaluation of spatial navigation cognitive ability of experimental animals and has good application effects.
