Objective To investigate the efficacy and mechanism of Qizhi Weitong Granule(QWG)in the treatment of ulcerative colitis.Methods A rat model of ulcerative colitis induced by 2,4,6-trinitrobenzene sulfonic acid was established.The therapeutic effect of QWG on ulcerative colitis(UC)rats was evaluated by the expressions of tight junction proteins Occludin and ZO-1.By means of 1H-NMR metabolomics technology and multivariate statistical analysis,the dynamic changes of endogenous metabolites were analyzed.And related metabolic pathways and metabolic networks were constructed to explore the possible mechanism of QWG in the treatment of ulcerative colitis.Results QWG had obvious repairing effect on colonic ulcer injury,and significantly increased the expression of Occludin and ZO-1 in colonic tissue(P<0.01).A total of 12 related metabolic biomarkers were screened and identified.Conclusion QWG has obvious protective effect on ulcerative colitis,and the main intervention pathways are pyruvate pathway and tricarboxylic acid cycle.
