Objective To explore the correlation between some metallic elements of Diaphragma Juglandis Fructus(DJF)from Yunnan and the protective effect of bilateral clamped renal ischemia-reperfusion injury(RIRI)in mice, and to provide the basis for clarifying the traditional medicinal material basis of distractor. Methods The deionized aqueous extract of DJF from Yunnan was prepared by using strong acid cation exchange resin. The contents of seven metallic elements in the deionized aqueous extract of DJF,as well as in aqueous extract of DJF and ethanol extract of DJF,were measured by inductively coupled plasma atomic emission spectrometry(ICP-AES).The mouse model of RIRI was established by bilateral clamping method, and the correlation between some metallic elements in DJF extracts and biochemical indexes of RIRI mice was analyzed. Results Every group of DJF extract could reduce the levels of serum creatinine(CRE)and urea nitrogen(BUN)in mice with RIRI,reducing IL-1β and TNF-α level. The contents of Mg and Zn in the extracts were(negatively)significantly correlated with the content of CRE of RIRI mouse model;the content of Mn was(negatively)significantly correlated with BUN and IL-1p.The expression level was negatively correlated with the content of Fe and TNF-α.The experimental results showed that some metallic elements in DJF had the protective effect on RIRI.Conclusion Some metallic elements of DJF are related to their role in preventing bilateral clamped RIRI in mice, and can protect RIRI in a certain range.
