创伤性脑损伤及其治疗药物

创伤性脑损伤(traumatic brain injury,TBI)是一种威胁人类生命安全的常见损伤。由于其损伤机制复杂,目前外科手术是临床首选治疗手段,但针对手术后继发性TBI的预防与治疗尚没有有效治疗药物。研究TBI损伤机制、寻找有效治疗药物以减轻患者痛苦、提高生活质量,成为目前的研究热点。本综述概括了近几年有关TBI治疗的最新进展,包括TBI损伤机制、TBI动物模型建立和评价指标、治疗药物及其新型递送系统,其中新型药物递送系统概括了聚合物纳米粒、水凝胶、树枝状聚合物、脂质体、胶束和细胞外囊泡作为新型药物载体在TBI治疗中的应用,希望为安全、有效地治疗TBI提供新思路。
Traumatic brain injury(TBI)and its therapeutic drugs
Traumatic brain injury(TBI)is a common injury that threatens the safety of human life.Due to its complex injury mechanism,only surgery is used as an emergency treatment method in clinics.However,it lacks the efficient and safe strategies for the prevention and protection of the secondary TBI.The research on TBI mechanism and the search for effective therapeutic drugs to alleviate the pain of patients have become the focus of current research and received extensive attention.This paper reviews the research progress of traumatic brain injury(TBI)in recent years,including injury mechanism,the establishment of animal experimental model and the evaluation index,potential therapeutic drugs and drug delivery systems.The related drug delivery systems summarize the applications of polymer nanoparticles,hydrogels,dendritic polymers,liposomes,micelles and extracellular vesicles as drug carriers in TBI therapy.
