Cinnamomum cassia Prsel extract via regulate COX-2 expression to alleviate benign prostatic hyperplasia in rats
Objective To investigate the therapeutic effect of Cinnamomum cassia Presl on benign prostatic hyperplasia(BPH)and its potential mechanism.Methods Molecular docking preliminarily predicted the binding ability of three compounds in CE to cyclooxygenase-2(COX-2)and 5-lipoxygenase(5-LOX).The prostate weight(PW)and prostate index(PI)were examined.The histomorphological changes of prostate were observed by hematoxylin eosin staining.The protein and mRNA expression of COX-2 and 5-LOX in rat prostate tissue were detected by Western-blot and q-PCR,respectively.Results The molecular docking showed that the three main active compounds in CE had good binding energy with COX-2 and 5-LOX protein models.The Western-blot and q-PCR showed that Cinnamomum cassia Presl could reduce the expression of COX-2 and 5-LOX at the protein and mRNA level at the same time,and the down-regulation of COX-2 was significant.Meanwhile,the results of PW,PI and histopathology also revealed that Cinnamomum cassia Presl significantly ameliorated BPH.Conclusion Cinnamomum cassia can effectively reverse BPH,and its mechanism may be related to down-regulating the expression of COX-2 in arachidonic acid metabolic network.
