基于"药物→ 疾病"逆向分析策略运用生物标签研究模式分析杜仲叶治疗血液疾病的作用机制及物质基础

目的 基于"药物→疾病"逆向分析策略运用生物标签研究模式分析杜仲叶治疗血液疾病的潜能、作用机制及物质基础.方法 运用蛋白质组学和代谢组学探索杜仲叶干预血清样本的生物标签,再运用生物信息学拓扑分析其治疗血液疾病的潜能、作用机制和物质基础.结果 6个差异表达蛋白质和6个差异表达代谢物参与了 3个显著调节通路,这些内源性靶点可以作为潜在生物标签.通过生物信息学数据库的分析,杜仲叶的治疗潜能可能涉及白细胞障碍、血小板病、白血病和贫血.杜仲叶的65个化学成分可能成为其治疗血液疾病的物质基础.结论 生物标签研究模式揭示了杜仲叶治疗血液疾病的潜能、作用机制及物质基础.可为后续将杜仲叶开发成保健食品,甚至成为药物提供重要的参考依据.
Applying biolabel-led research pattern to analyze the mechanism of action and material basis of eucommiae folium in the treatment of blood diseases based on"drug →disease"reverse analysis strategy
Objective Biolabel-led research pattern was used to analyze the therapeutic potential,mechanism of action,and material basis of eucommiae folium(EF)in the treatment of blood diseases by using biolabel-led research pattern based on"drug → disease"reverse analysis strategy.Methods Proteomics and metabonomics were applied to explore the biolabels responsible for the effect of EF on serum samples.Subsequently,bioinformatics was used to analyze its therapeutic potential,mechanism of action,and material basis.Results Omics analysis showed that six proteins and six metabolites were differentially expressed,which were involved in three significantly dysregulated pathways and could be used as potential biolabels.Bioinformatics analysis showed that the therapeutic potential of EF may be involved in leukocyte disorders,thrombocytopathy,leukemia,and anemia.Sixty-five compounds from EF may be the material basis for the treatment of blood diseases.Conclusion Biolabel-led research pattern reveals the therapeutic potential,mechanism of action,and material basis of EF in the treatment of blood diseases.This can provide an important reference for the development of EF into health food and even drug.
