基于Box-Behnken响应面法优化膝痛康β-蜕皮甾酮的提取工艺

目的 优化膝痛康方剂中β-蜕皮甾酮的提取工艺。方法 采用Box-Behnken响应面法和单因素试验,对提取过程中的浸泡时间、提取次数、提取时间和甲醇倍量等进行优化,通过响应面模型分析与预测,得到最佳的提取工艺条件,并对最优条件进行验证。结果 饮片浸泡96 min,提取100 min,反复2次,甲醇倍数为16倍,此时牛膝的提取工艺达到最佳,在此提取工艺下β-蜕皮甾酮的提取量达到最大。结论 该提取方法具有设备简单、成本低、提取工艺稳定等特点,此外,该提取工艺具有较好的应用价值,以其为膝痛康的应用提供实验依据。
Optimization of extraction process of β-ecdysterone from Xitongkang based on Box-Behnken response surface methodology
Objective To optimize the extraction process of β-ecdysterone in Xitongkang formula.Methods The soaking time,extraction times,extraction time and methanol multiplicity in the extraction process were optimized by using one-way test and Box-Behnken response surface method,and the optimal extraction process conditions were obtained through response surface model analysis and prediction,and the optimal conditions were verified.Results The optimal extraction process of Cynodon grandiflorus was as follows:the slices was soak for 96 min and extracted for 2 times,100 min for each 16 times of methanol were used.Conclusion The extraction method was characterized by simple equipment,low cost and stable extraction process.Moreover,the extraction process has good application value which could provide theoretical basis for the application of Xitongkang.
