细胞色素P450-RhFRED人工融合酶的研究进展与应用

来自于红球菌Rhodococcus sp。NCIMB 9784的P450RhF是一个自给自足型的细胞色素P450酶,它的血红素结构域主要负责控制酶的底物特异性,P450还原酶伴侣蛋白RhFRED与细胞色素P450血红素结构域天然融合在一起,电子主要从蛋白分子内转移到P450血红素结构域。依据融合型P450RhF自给自足的特性,有学者利用P450RhF的还原伴侣蛋白RhFRED,与其它P450构建人工融合酶P450-RhFRED,来实现多种细胞色素P450基因的功能表达。基于此建立的P450-Rh-FRED融合文库对未知功能P450的催化功能探索有很大意义,P450-RhFRED融合酶技术也可用于开发已知细胞色素P450酶的工业应用。
Research progress and application of cytochrome P450-RhFRED artificial fusion enzyme
P450RhF from Rhodococcus sp.NCIMB 9784 is a self-sufficient cytochrome P450 enzyme,whose heme domain is mainly responsible for controlling the substrate specificity of the enzyme,the redox partner RhFRED naturally fuses with the P450 heme domain.Electrons were transferred within the protein molecule.Based on the self-sufficiency of P450RhF,some scholars successively constructed artificial fusion enzyme P450-RhFRED with other P450s by using the redox partner RhFRED of P450RhF to construct functional P450 enzymes.The P450-RhFRED fusion protein library is of great significance for the exploration of the catalytic function of unknown P450s.P450-RhFRED artificial fusion proteins can also be used in development of enzymes for industrial applications.
