Optimization of extraction process for deer keratin based on Box-Behnken response surface methodology
Objective To optimize the extraction process of antler protein by Box-Behnken response surface method.Methods The response surface method was designed on the basis of univariate investigation to find the best extraction conditions.The protein content in the antler was used as the investigation index,the absorbance of the sample solution was determined,and the protein content of the sample was calculated according to the standard curve equation.Results The optimal extraction process conditions were as follows:NaOH concentration was 0.35 mol·L-1,liquid-to-material ratio was 6:1,extraction time was 114 min.Conclusion It is verified that the deviation between the actual measured value and the predicted value obtained by the Box-Behnken response surface method is within±1%,and the detection results are good,which can provide a reference for the extraction process of antler protein in Xitongkang.
