The effects of the protective coat types of metoprolol succinate sustained-release pellets on the formation of the pellet-based tablets:An initial evaluation
Objective To evaluate the mechanical protective effects of hydroxypropyl cellulose(HPC)and hydroxypropyl methylcellulose(HPMC)on metoprolol succinate sustained-release pellets.Methods Metoprolol succinate sustained-release pellets were coated with different grades of HPC and HPMC.The protective effects of these polymer materials were evaluated by pressure load,integrity,and in vitro dissolution.HPC and HPMC films were prepared and their mechanical properties were investigated.Results Metoprolol succinate pellets coated with HPC and HPMC increased the fracture load and mechanical properties,improved integrity,and had a lower dose dumping.Conclusion HPC and HPMC protective coating layer have certain mechanical protective effects on the release units of metoprolol succinate tablets and HPC has relatively higher protective ability.This research may provide reference for the pellet protection strategy of multi-unit pellet drug release system.
