Determination of content component amino acids in compound glycyrrhizin preparation by derivatization of succinimide benzoatechang
Objective To establish a method for the analysis of amino acids in Compound Glycyrrhizin preparations derived from succinimide benzoate.Methods Succinimide benzoate was used as the derivatization reagent.The separation was performed on Angla innovation C18 column(150 mm×4.6 mm,5 μm),methanol as mobile phase A,0.5%triethylamine phosphate buffer(pH 5.00)as mobile phase B with gradient elution.The detection wavelength was 236 nm,the injection volume was 10 μL,the column temperature was room temperature,and the flow rate was 1.0 mL·min-1.Results Glycine and methionine in Compound Glycyrrhizin preparation had the good linear relationship in the range of 0.501-50.1 mg·L-1 and 1.00-50.2 mg·L-1 respectively,r=0.999 6,r=0.999 7;The recovery rate met the requirements of content component analysis in preparation analysis.Conclusion The method reduces the operation error caused by multiple dilution of content components in the analysis of high-sensitivity derivative reagents,but also does not interfere with the separation after hydrolysis of excessive derivative reagents.The method is simple,fast and accurate.It can be used as an analysis method of constant amino acids in pharmaceutical preparations.
