LncRNA ITGB1在垂体瘤中的表达及对细胞生物学行为的影响

目的 检测长链非编码RNA(LncRNA)ITGB1 在垂体瘤中的表达并探讨其对垂体瘤细胞增殖、凋亡、侵袭、迁移行为的影响。方法 选取2018 年9 月—2021 年4 月在本院接受手术治疗的48例垂体瘤患者及死于意外的48 例组织捐献者,分别收集垂体瘤及正常垂体前叶组织。取人垂体瘤RC-4BC细胞,分别构建 ITGB1 敲低(转染 LncRNA ITGB1 siRNA)和过表达株(转染 pcD-NA3。1 LncRNA ITGB1),qPCR法检测RC-4BC细胞中LncRNA ITGB1 表达,Western Blot法检测增殖细胞核抗原(PCNA)、含半胱氨酸的天冬氨酸蛋白水解酶 3(Caspase-3)、基质金属蛋白酶-9(MMP-9)蛋白表达。结果 垂体瘤组织中LncRNA ITGB1 表达水平较正常垂体前叶组织升高;Ln-cRNA ITGB1 在不同肿瘤大小、侵袭性患者垂体瘤组织中表达差异具有统计学意义;与siRNA NC组相比,LncRNA ITGB1 siRNA组RC-4BC细胞增殖率、侵袭数、划痕愈合率及细胞中LncRNA IT-GB1 水平、PCNA、MMP-9 蛋白水平降低,RC-4BC细胞凋亡率及细胞中Caspase-3 蛋白水平升高;与pcDNA3。1 NC组相比,pcDNA3。1 LncRNA ITGB1 组RC-4BC细胞增殖率、侵袭数、划痕愈合率及细胞中 LncRNA ITGB1 水平、PCNA、MMP-9 蛋白水平升高,RC-4BC 细胞凋亡率及细胞中Caspase-3 蛋白水平降低。结论 LncRNA ITGB1 在垂体瘤中表达上调,与垂体瘤患者肿瘤大小、侵袭性有关,可促进垂体瘤RC-4BC细胞的增殖、侵袭、迁移过程,并抑制细胞凋亡。
Expression of LncRNA ITGB1 in pituitary tumors and its influence on cell biological behavior
Objective To detect the expression of long non-coding RNA(LncRNA)ITGB1 in pituitary tumors and to explore its effects on the proliferation,apoptosis,invasion and migration behavior of pituitary tumor cells.Methods Forty-eight patients with pituitary tumors and 48 tissue donors who died of accidents who underwent surgical treatment in our hospital from September 2018 to April 2021 were selected to collect pituitary tumor tissue and normal anterior pituitary tissue respectively.Human pituitary tumor RC-4BC cells were divided into control group,siRNA NC group(transfected with nonsense siRNA),LncRNA ITGB1 siRNA group(transfected with LncRNA ITGB1 siRNA),pcDNA3.1 NC group(transfected with pcDNA3.1)and pcDNA3.1 LncRNA ITGB1 group(transfected with pcDNA3.1 LncRNA ITGB1),qPCR method was used to detect the expression of LncRNA ITGB1 in RC-4BC cells,Western Blot method was used to detect the protein expression of proliferating cell nuclear antigen(PCNA),cysteinyl aspartate specific proteinase-3(Caspase-3)and matrix metalloproteinase-9(MMP-9).Results The expression level of LncRNA ITGB1 in pituitary tumor tissue was higher than that in normal anterior pituitary tissue;The expression of LncRNA ITGB1 in pituitary tumor tissues of different tumor size and invasive patients was different statistically;compared with the siRNA NC group,the RC-4BC cell proliferation rate,invasion number,scratch healing rate,LncRNA ITGB1 level,PCNA and MMP-9 protein levels in the LncRNA ITGB1 siRNA group were reduced,the apoptotic rate of RC-4BC cells and the level of Caspase-3 protein in the cells were increased;compared with pcDNA3.1 NC group,the RC-4BC cell proliferation rate,invasion number,scratch healing rate,LncRNA ITGB1 level,PCNA and MMP-9 protein levels in the pcDNA3.1 LncRNA ITGB1 group were increased,the apoptotic rate of RC-4BC cells and the level of Caspase-3 protein in the cells were reduced.Conclusion LncRNA ITGB1 is up-regulated in pituitary tumor,and related to the tumor size and invasion in patients with pituitary tumor,it can promote the proliferation,invasion and migration of pituitary tumor RC-4BC cells,and inhibit cell apoptosis.
