The effect of glucagon-like peptide 2 receptor gene on prognosis and immune infiltration of liver cancer
Objective To investigate the differential expression of glucagon like peptide 2 receptor(GLP2R)in hepatocellular carcinoma(HCC)tissues and normal liver tissues using bioinformatics methods and to explore its role in prognosis and immune cell infiltration in patients with liver cancer.Methods TIMER2.0 database was used to investigate the expression difference of GLP2R in various tumor tissues and its corresponding normal tissues.The hepatocellular carcinoma data sets of TCGA database was downloaded,and R software was used to explore the expression difference of GLP2R in liver cancer tissues and normal tissues.Kaplan-Meier survival curve was drawn to analyze the relationship between GLP2R expression level and HCC prognosis.R software was used to analyze the liver cancer data sets to obtain differential genes.The interaction network between GLP2R and differential genes was constructed using STRING and Cytoscape to obtain key genes.GLP2R and key genes were analyzed by univariate and multivariate Cox regression analysis.The enrichment pathway of GLP2R and key genes was analyzed by GO.TIMER2.0 database was used to explore the correlation between GLP2R and immune cell infiltration in TIMER and CIBERSORT.CIBERSORT was used to further analyze the differences of infiltrating immune cells in liver cancer.Results Compared with normal tissues,GLP2R mRNA expression level in liver cancer tissues was down-regulated.Liver cancer patients with low GLP2R expression had better prognosis.GLP2R could be used as an independent predictor of poor prognosis in liver cancer patients.GO enrichment analysis showed that GLP2R-related genes were mainly enriched in biological processes such as cell response to glucagon stimulation.The expression of GLP2R in HCC was negatively correlated with the immunoinfiltration level of CD8+ T cells.Conclusions GLP2R expression is low in liver cancer tissues.Low expression of GLP2R is associated with better prognosis of liver cancer patients and affects the level of immune infiltration of HCC.GLP2R gene can be used as a potential immunotherapeutic target for HCC.
