Dalbergiphenol induces autophagy and apoptosis of bladder cancer UMUC3 cells
Objective To investigate the effect of dalbergiphenol(DAL),a flavonoids extracted from Indian sandalwood,on autophagy and apoptosis of bladder cancer cells and its potential molecular mechanisms.Methods Bladder cancer cell line UMUC3 were cultured.After treatment of the cells with various concentrations(0,10,20,40 and 80 μM)of DAL,the inhibition effect of the compound on the cells were detected by CCK-8 test.The effect of DAL on the migration was detected by cell scratch assay.The effect of DAL on apoptosis was examined by using TUNEL assays.The autophagy and AKT/mTOR signaling pathway related proteins,including LC3-II,p62,p-PI3K,p-Akt,p-mTOR,AKT and mTOR were detected by Western blot.Results Compared to blank group(DAL concentration=0),DAL inhibited proliferation,migration,colony-formation and induction of apoptosis of UMUC3 cells in a dose-dependent manner(P<0.05).Western blot results showed that expressions of LC3-Ⅱ/LC3-Ⅰ and Beclin-1 were obviously increased while P62 was decreased in the DAL treatment cells when compared to the blank group(P<0.05).The expressions of p-Akt and phosphorylated t-mTOR protein were significantly decreased in the DAL treatment cells when compared to the blank group(P<0.05).However,DAL had no significant effect on protein expressions of t-Akt and t-mTOR(P>0.05).Conclusions DAL could induce the autophagy and apoptosis of UMUC3 cells.Its mechanism may through activating the AKT/mTOR signaling pathway.
