摘要
目的 探讨miR-379-5p及miR-1255b-5p在肺癌组织的表达情况及意义.方法 我院经手术治疗且经组织病理学确诊的肺癌患者96 例,术中切除肺癌组织及癌旁组织后液氮速冻-80℃留存,采用实时荧光定量反转录聚合酶链反应测定组织中miR-379-5p及miR-1255b-5p表达情况,分析二者检测结果与肺癌病理指标、生存预后的关系.结果 相较于癌旁组织,肺癌组织中的miR-379-5p、miR-1255b-5p相对表达量均显著降低(P<0.05);肺癌患者癌组织中的miR-379-5p、miR-1255b-5p相对表达量呈正相关(r=0.759,P<0.05);对比临床分期Ⅰ~Ⅱ期者,Ⅲ期者癌组织中miR-379-5p、miR-1255b-5p相对表达量显著更低(P<0.05);对比无淋巴结转移者,有淋巴结转移者癌组织中miR-379-5p、miR-1255b-5p相对表达量显著更低(P<0.05);以miR-379-5p(均值0.43)、miR-1255b-5p(均值0.51)相对表达量的均值为界分为低表达和高表达患者,随访截止至2023年6月,miR-379-5p低表达者(51 例)的生存期短于miR-379-5p高表达者(45 例),miR-1255b-5p低表达者(47 例)的生存期短于miR-1255b-5p高表达者(49 例)(P<0.05);临床分期Ⅲ期、淋巴结转移、miR-379-5p低表达、miR-1255b-5p低表达是影响肺癌患者生存期的危险因素(P<0.05).结论 肺癌患者癌组织中miR-379-5p及miR-1255b-5p相对表达量异常降低,且与临床分期、淋巴结转移及生存预后有关.
Abstract
Objective To explore the expression changes and significance of miR-379-5p and miR-1255b-5p in lung cancer tissues.Methods Totally 96 patients with lung cancer who received surgical treatment and were confirmed by histopathology in our hospital were selected.The lung cancer tissues and adjacent tissues were surgically removed and preserved at-80℃by liquid nitrogen quick-freezing.The expressions of miR-379-5p and miR-1255b-5p in tissues were detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction.The relationship between the detection results of both indicators and pathological indicators and survival prognosis of lung cancer was analyzed.Results Compared with adjacent tissues,the relative expression levels of miR-379-5p and miR-1255b-5p in lung cancer tissues were significantly decreased(P<0.05).The relative expression levels of miR-379-5p and miR-1255b-5p in cancer tissues of patients with lung cancer were positively correlated with each other(r=0.759,P<0.05).The relative expression levels of miR-379-5p and miR-1255b-5p in cancer tissues were significantly lower in patients with clinical stage Ⅲ when compared with those in patients with clinical stage Ⅰ to Ⅱ(P<0.05).Compared with patients without lymph node metastasis,the relative expression levels of miR-379-5p and miR-1255b-5p in cancer tissues of patients with lymph node metastasis were obviously lower(P<0.05).The patients were divided into low expression group and high expression group based on the mean relative expression levels of miR-379-5p(mean=0.43)and miR-1255b-5p(mean=0.51).Follow-up until June 2023,51 patients with low expression of miR-379-5p had shorter survival time when compared to 45 patients with high miR-379-5p expression.The survival time of 47 patients with low miR-1255b-5p expression was shorter than that of 49 patients with high miR-1255b-5p expression(P<0.05).Clinical stage Ⅲ,lymph node metastasis,low expression of miR-379-5p,and low expression of miR-1255b-5p were risk factors affecting the survival time in patients with lung cancer(P<0.05).Conclusions The relative expression levels of miR-379-5p and miR-1255b-5p in cancer tissues of patients with lung cancer are abnormally decreased.Their ex-pressions are correlated with clinical staging,lymph node metastasis and survival prognosis.
