Study on Mechanism of Shengjiang Longlian Decoction(升降龙莲汤)in the Treatment of Endometrial cancer Based on Network Pharmacology,GEO database and Molecular Docking
Objective This study was based on network pharmacology,gene chip and molecular docking to explore the mecha-nism of the treatment of endometrial cancer with empirical prescription,Shengjiang Longlian Decoction(升降龙莲汤).Methods The effective components of Shengjiang Longlian Decoction were extracted from TCMSP database by oral bioavailability(OB)and drug-likeness(DL),and target genes of endometrial cancer were obtained by GSE115810 dataset in GEO,OMIM and GeneCards database.Common targets of active ingredients and endometrial cancer were analyzed for GO function and KEGG pathway enrichment.The protein interaction PPI network was established using STRING,and core targets were screened accord-ing to the degree value after being introduced into Cytoscape.Then,the up-regulation and down-regulation of differential genes were integrated,and the target genes that were more closely related to the prognosis of endometrial cancer were further screened by Kaplan Meier-Plotter survival curve analysis.Finally,AutoDock was used for molecular docking verification of key active ingre-dients and core target genes.Results 66 active components and 289 potential therapeutic targets were obtained from Shengjiang Longlian Decoction.A total of 1048 targets related to endometrial cancer were obtained through three data platforms,including 97 intersection targets.Twelve target genes were obtained from PPI network,and seven were further screened by K-M survival curve.GO and KEGG enrichment results showed that the positive regulation of cell migration was the most significant in biologi-cal processes,and cancer signaling pathways were the most enriched.The molecular docking verification showed that quercetin,luteolin,wogonin,kaempferol and baicalein were closely bound to target genes.Conclusion The main mechanism of Shengjiang Longlian Decoction in endometrial cancer is to inhibit or improve the expression of key genes,block the proliferation and invasion of cancer cells,and promote the apoptosis of tumor cells through multi-target and multi-signal pathways.
