基于网络药理学和动物实验探究高优-巴沙嘎水提物对乳腺增生的影响

目的 通过网络药理学和实验验证并分析高优-巴沙嘎水提物(DSWE)治疗乳腺增生作用机制.方法 通过TCMSP和Swiss Target Prediction数据库检索DSWE相关靶点;在GeneCards、NCBI及OMIM数据库收集乳腺增生相关基因;构建PPI网络并对其共有靶点进行GO和KEGG富集分析.构建Wistar大鼠乳腺增生模型,采用Elisa法检测DSWE干预前后大鼠血清中雌二醇和孕酮的表达水平.结果 通过构建成分-疾病-通路-靶点网络,提示DSWE中存在6种活性成分可通过多条通路抑制乳腺增生.高剂量组DSWE可以显著降低雌二醇的表达,增加孕酮的表达.结论 DSWE能够通过多成分-多靶点-多通路发挥抑制乳腺增生的作用,这将为DSWE临床应用治疗乳腺增生提供初步实验数据支持,但更深入的分子机制仍然需要进一步探讨.
Exploring the Effect of Gaoyou Bashaga Water Extract on Breast Hyperplasia Based on Network Pharmacology and Experimental Validation
Objective It is to verify and analyze the mechanism of Gaoyou-Bashaga water extract(DSWE)in the treatment of mammary gland hyperplasia through network pharmacology and experiments.Methods DSWE related targets were searched through TCMSP and Swiss Target Prediction databases.Breast hyperplasia related genes were collected from GeneCards,NCBI and OMIM databases.PPI network was constructed and performed for their common targets.The Wistar rat mammary hyperplasia mod-el was constructed to determine the expression levels of estradiol and progesterone in serum with DSWE intervention by Elisa.Re-sults By constructing a component-disease-pathway-target network,it shows that six active components in DSWE can inhibit breast hyperplasia through multiple pathways.DSWE in the high-dose group could significantly reduce estradiol expression and increase the expression of progesterone.Conclusion DSWE can inhibit mammary gland hyperplasia through multi-component-multi-target-multi-pathway,which will provide preliminary experimental data support for the clinical application of DSWE in the treatment of mammary gland hyperplasia,but the in-depth molecular mechanism still needs to be further explored.
