2型糖尿病合并脑梗死的生信研究及潜在中药预测

目的 基于生物信息学的方法探索2型糖尿病(Diabetes Meditus type2,T2DM)合并脑梗死的关键基因和潜在的有效中药。方法 从GEO数据库下载T2DM基因芯片数据GSE29221以及脑梗死芯片数据GSE97537,确定差异基因(DEGs),在Metascape数据库完成交集DEGs的GO和KEGG分析,并完成蛋白质互作网络(PPI)分析,并通过Cytoscape-v3。8进行可视化分析。运用CoremineMedical检索平台,筛选出治疗T2DM合并脑梗死的潜在中药。结果 根据筛选条件得出,GSE29221芯片中共包括1445个DEGs(1156个上调基因、289个下调基因);GSE97537芯片含有DEGs10842个(3884个上调基因、4080个下调基因)。Metascape在线分析显示DEGs的生物学过程(BP)主要涉及调节平滑肌细胞增殖、正向调节蛋白质磷酸化及程序性细胞死亡等;细胞组分(CC)主要包含细胞外基质、囊泡腔等;分子功能(MF)主要影响细胞因子活性、受体配体活性、生长因子活性等。同时,KEGG通路富集分析显示T2DM合并脑梗死可能与AGE-RAGE、PI3K-Akt、Foxo、Jak-STAT、MAPK等信号通路或联级有关,同时可能涉及HIF-1信号、流体剪切应力和动脉粥样硬化、内分泌抵抗、细胞凋亡、长寿调节途径、胰岛素抵抗等生物学过程。经CoremineMedical分析得出黄芪、川芎、人参、红花、丹参、赤芍、生姜、桃仁、茯苓、当归与关键基因关系较为密切。结论 T2DM合并脑梗死的发病可能涉及多通路、多靶点的复杂机制,黄芪、川芎、人参等是与T2DM合并脑梗死的关键基因最密切的中药。
Bioinformatics Research and Potential Chinese Medicine Prediction of Type 2 Diabetes Mellitus Complicated with Cerebral Infarction
Objective Based on bioinformatics methods,the key genes and potential effective traditional Chinese medicines of type 2 diabetes mellitus(T2DM)with cerebral infarction were explored.Methods T2DM gene chip data GSE29221 and cerebral infarction chip data GSE97537 were downloaded from GEO database to determine differential genes(DEGs).GO and KEGG a-nalysis of intersection DEGs were completed in Metascape database,and protein interaction network(PPI)analysis was comple-ted,and visual analysis was performed by Cytoscape-v3.8.Coremine Medical retrieval platform was used to screen out the po-tential Chinese medicines for the treatment of T2DM with cerebral infarction.Results According to the screening conditions,the GSE29221 chip contained 1445 DEGs(1156 up-regulated genes and 289 down-regulated genes).The GSE97537 chip con-tains 10842 DEGs(3884 up-regulated genes and 4080 down-regulated genes).Metascape online analysis showed that the bio-logical processes(BP)of DEGs were mainly involved in regulating smooth muscle cell proliferation,positively regulating protein phosphorylation and programmed cell death.Cell component(CC)mainly included extracellular matrix,vesicle cavity,etc.Mo-lecular function(MF)mainly affected cytokine activity,receptor ligand activity,growth factor activity,etc.At the same time,KEGG pathway enrichment analysis showed that T2DM complicated with cerebral infarction may be related to AGE-RAGE,PI3K-Akt,Foxo,Jak-STAT,MAPK and other signaling pathways or cascades,and may also involve H1F-1 signaling,fluid shear stress and atherosclerosis,endocrine resistance,apoptosis,longevity regulation pathway,insulin resistance and other biologi-cal processes.Coremine Medical analysis showed that Huangqi(Astragali Radix),Chuanxiong(Chuanxiong Rhizoma),Renshen(Ginseng Radix Et Rhizoma),Honghua(Carthami Flos),Danshen(Salviae Miltiorrhizae Radix Et Rhizoma),Chishao(Paeoniae Radix Rubra),Shengjiang(Zingiberis Rhizoma Recens),Taoren(Persicae Semen),Fuling(Poria)and Danggui(Angelicae Sinensis Radix)were closely related to key genes.Conclusion The pathogenesis of T2DM complicated with cerebral infarction may involve a complex mechanism of multiple pathways and multiple targets.Huangqi(Astragali Radix),Chuanxiong(Chuanx-iong Rhizoma)and Renshen(Ginseng Radix Et Rhizoma)are most closely related to the key genes of T2DM complicated with cerebral infarction.
