射干药效物质基础成分鸢尾黄素、鸢尾甲黄素A的pKa值测定

目的 采用高效液相色谱法测定鸢尾黄素、鸢尾甲黄素A的pKa值,为其剂型设计提供参考.方法 采用乙腈-不同pH值磷酸缓冲盐水溶液为流动相,在检测波长265 nm下,依次测定鸢尾黄素、鸢尾甲黄素A的保留因子,从而计算pKa.结果 鸢尾黄素的pKa为6.49、鸢尾甲黄素A的pKa为6.51.结论 采用高效液相色谱法测定鸢尾黄素、鸢尾甲黄素A的pKa值,方法简便、有效.
Determination of pKa Value of Pharmacodynamic Substance Basis Tectorigenin and Iristectorigenin A of Shegan(Belamcanda Chinensis)
Objective To determine the pKa values of tectorigenin and iristectorigenin A by high performance liquid chromatog-raphy and provide reference for the formulation design.Methods The mobile phase was acetonitrile phosphoric acid buffer solution with different pH values.At the detection wavelength of 265 nm,the retention factors of tectorigenin and iristectorigenin A were determined successively and then the pKa value was calculated.Results The pKa values of tectorigenin was 6.49 and that of iris-tectorigenin A was 6.51.Conclusion High performance liquid chromatography is a simple and effective method for the determina-tion of pKa value of tectorigenin and iristectorigenin A.
