Anti-diabetic Activity of Sanqi(Panax notoginseng)Leaves Based on Network Pharmacology and Diabetic Rats
Objective The main active ingredients and potential mechanisms of action of Sanqi(Panax notoginseng)leaves a-gainst diabetes were investigated using network pharmacology and animal experiments. Methods After screening the potential tar-gets shared by Sanqi(Panax notoginseng)leaves and diabetes based on Pubchem,PharmMapper and OMIM databases. The STRING database and Cytoscape software were used to construct protein interaction network maps. The topological analysis and cluster analysis were performed to obtain the core targets, and GO and KEGG enrichment analyses were performed to screen the possible mechanisms of drug action on the disease.Subsequently, animal experiments were conducted to verify the antidiabetic effect of Sanqi(Panax notoginseng)leaves. Results A total of 50 common targets of Sanqi(Panax notoginseng)leaves and diabe-tes were obtained, with three core targets:ANXA5,CFB and GSTP1,involving 986 biological processes,15 cellular compositions,74 molecular functions and 101 signaling pathways.Animal experiments have shown that Sanqi(Panax notoginseng)leaves can decrease blood glucose in diabetic rats, reduce the expressions of p38 MAPK and TGF-β1 mRNA associated with diabetic ne-phropathy,as well as regulate oxidative stress and lipid levels and reduce atherosclerosis. Conclusion Sanqi(Panax notoginseng)leaves exert its anti-diabetic effects through a combination of multiple pathways and targets, and its mechanism of action may be related to reducing oxidative stress, regulating cytokines, reducing atherosclerosis and anti-diabetic nephropathy renal fibrosis.
