自拟桃仁白芍三七粉对黄褐斑模型小鼠的治疗作用研究

目的 探讨自拟桃仁白芍三七粉对黄褐斑模型小鼠的治疗作用及其作用机制。方法 将100只小鼠随机分为空白组,模型组,桃仁低、高剂量组,白芍低、高剂量组,三七低、高剂量组,桃仁白芍三七粉低、高剂量组,共10组,每组10只。采用黄体酮注射液联合中波紫外线照射的方法制备黄褐斑模型小鼠,测定皮肤和肝脏组织中超氧化物歧化酶(SOD)活性以及丙二醛(MDA)、酪氨酸酶(TYR)水平,并进行皮肤病理学观察。结果 与模型组相比,桃仁白芍三七粉低、高剂量组均可显著提高皮肤和肝脏组织中的SOD活性(P<0。05或P<0。01),可以显著降低MDA及TYR的含量(P<0。05或P<0。01)。皮肤病理学实验结果显示:小鼠皮肤中黑色素细胞沉着减轻、黑色素细胞个数显著减少(P<0。05或P<0。01),且高剂量组与各单药组相比,表现出一定的协同增效作用。桃仁白芍三七粉低、高剂量组可以改善小鼠表皮角质化以及上皮细胞的增生现象,真皮层的皮肤纤维结构、毛囊以及皮脂腺、皮下组织炎症浸润情况与模型组相比也有明显的改善。结论 自拟桃仁白芍三七粉有一定的治疗黄褐斑的功效,其作用与提高机体抗氧化能力,抑制酪氨酸酶活性,改善机体的炎症有关。
Study on the Therapeutic Effects of Self-Made Persicae Semen & Paeoniae Radix Alba & Panax Notoginseng powder on Melasma Model Mice
Objective To investigate the therapeutic effects and mechanism of self-made Persicae Semen & Paeoniae Radix Alba & Panax Notoginseng powder on melasma model mice.Methods 100 mice were randomly divided into blank group,model group,Persicae Semen low/high-dose group,Paeoniae Radix Alba low/high-dose group,Panax Notoginseng low/high-dose group,Persicae Semen & Paeoniae Radix Alba & Panax Notoginseng powder low/high-dose group,10 groups in total,10 mice in each group.Mice models of melasma were prepared by progesterone injection combined with UVA irradiation.Superoxide dis-mutase(SOD)activity,malondialdehyde(MDA),and tyrosinase(TYR)levels in skin and liver tissues were measured and dermatopathological observations were made.Results Compared with the model group,the activity of SOD in skin and liver tissue was significantly increased(P<0.05 or P<0.01),and the contents of MDA and TYR were significantly decreased(P<0.05 or P<0.01)in both low and high dose groups.The results of the skin pathology test showed that the melanocytes in the skin of mice were relieved and the number of melanocytes was significantly reduced(P<0.05 or P<0.01),and the high-dose group showed a certain synergistic effect compared with each single drug group.The epidermal keratinization and epithelial cell hyperplasia of mice were improved in the low and high-dose groups of peach kernel Paeonia Panax Notoginseng powder,and the skin fiber structure,hair follicles,sebaceous glands and inflammatory infiltration of subcutaneous tissue in the dermis were also significantly improved compared with the model group.Conclusion Self-made Persicae Semen & Paeoniae Radix Alba &Panax Notoginseng powder is effective in the treatment of melasma,which is related to improving the body's antioxidant capacity,inhibiting tyrosinase activity,and improving inflammation.
