Mechanism of Acupoint Thread Embedding Regulating NLRP3 Inflammasome to Protect Acute Renal Injury Model Rats Induced by Contrast Agents
;Objective To observe the mechanism of acupoint catgut embedding regulating the NLRP3 inflammasome signaling pathway in protecting renal function in contrast induced nephropathy rats.Method Sprague Dawley(SD)rats were randomly di-vided into blank group,model group and acupoint embedding group,with 10 rats in each group.Indomethacin,L-nitroarginine methyl ester(L-NAME)and iohexol injection were used to establish the rat model of contrast nephropathy.The acupoint em-bedding group was given simple embedding treatment.The pathological changes of rat kidney tissue were compared.The levels of IL-1 β,IL-10 and TNF-α in BALF in in serum and kidney tissue detected by enzyme-linked immunosorbent assay(ELISA)were compared.Western blot was used to detect the expressions of NLRP3 pathway related proteins.Results Compared with the model group,the score of renal function damage in the acupoint catgut embedding group was reduced,the inflammatory infiltration in the acupoint catgut embedding group was significantly reduced by HE staining,and the levels of IL-1 β,IL-10 and TNF-αin the serum and tissue of the kidney in the model group were significantly increased,while in the acupoint catgut embedding group,there was a significant decrease.At the same time,the differences in Western Blot detection were statistically significant(P<0.05).Conclusion Acupoint catgut therapy can significantly improve kidney inflammation in contrast nephropathy model rats,and reduce the levels of IL-1β,IL-10 TNF-α and histopathological scores.The mechanism of action may be related to the regulation of NLRP3 inflammasome signaling pathway.
