Molecular mechanism of Huanglian in the treatment of polycystic ovary syndrome with type 2 diabetes mellitus based on network pharmacology
Objective To explore the molecular mechanism of Huanglian in the treatment of polycystic ovary syndrome(P-COS)with type 2 diabetes mellitus(T2DM)through network pharmacology.Method the traditional Chinese medicine system pharmacology database TCMSP was used to obtain the information of effective components and target proteins of Huanglian.Uni-Prot database was used to convert the target names into common gene names.The genes related to PCOS and T2DM were screened by GeneCards database and matched with the corresponding gene names of the target of Huanglian.The network of Huanglian-components-targets was constructed by using Cytoscape 3.8.0 software,and the related gene enrichment was analyzed by using the database of functional annotation bioinformatics analysis platform(David).Results 14 active components of Huanglian were selected.There were 127 target genes predicted that Huanglian could treat PCOS and T2DM,which were significantly enriched in KEGG pathways such as thyroid hormone signaling pathway,inflammatory bowel disease,insulin signaling pathway and adipocyto-kine signaling pathway.AKT1,IL-6,TP53,VEGFA and TNF were the top five target genes of Huanglian.Conclusion PCOS and T2DM have common pathogenic genes.Huanglian can treat PCOS complicated with T2DM through multiple genes and pathways.
