Objective To explore the molecular mechanism of Bushen Huanjing Formula(补肾还精方)in improving the pre-mature ovarian failure caused by cyclophosphamide in mice.Method Sixty 10-week-old female C57/BL6 mice were randomly divided into five groups,with 12 mice in each group,namely the blank group,the model group,the Bushen Huanjing Formula low-dose group,the Bushen Huanjing Formula medium-dose group and the Bushen Huanjing Formula high-dose group.The mice were injected with cyclophosphamide intraperitoneally to establish a model,and were treated with different concentrations of Bushen Huanjing Formula by gavage.H&E staining was used to detect the pathological morphology of mouse ovary and count fol-licles at various types.The qPCR method was used to detect mRNA expression levels of cytochrome P450 family related proteins and cell senescent apoptotic protein genes.Results Compared with that in the model group,the ovarian atrophy in mice treated with drugs improved,with loose interstitial tissue and significantly reduced atretic follicles(P＜0.01).Compared with those in the model group,the mRNA expression levels of cytochrome P450 family genes were significantly increased(P＜0.05),while the mRNA expression levels of apoptosis-related proteins were significantly decreased(P＜0.05).Conclusion By enhancing the ex-pressions of P450 family-related proteins and inhibiting the expressions of senescence-associated apoptosis proteins,Bushen Huanjing Formula alleviates the injury of ovarian granulosa cells induced by cyclophosphamide and the production of atretic folli-cles to realize the therapeutic effect on premature ovarian failure mice.
