补肾还精方促进细胞色素P450基因表达改善卵巢早衰的机制研究

目的 探讨补肾还精方改善环磷酰胺所致小鼠卵巢早衰的分子机制。方法 将60只10周龄的C57/BL6雌性小鼠,随机分成5组,每组12只,分别为空白组、模型组、补肾还精方低剂量组、中剂量组、高剂量组,并利用环磷酰胺腹腔注射造模,并施加不同浓度的补肾还精方灌胃干预。采用H&E染色法观察小鼠卵巢病理形态,并进行各级卵泡计数;qRT-PCR法检测细胞色素P450家族相关蛋白和细胞衰老凋亡蛋白基因mRNA表达水平。结果 与模型组比较,通过中药干预后的小鼠卵巢萎缩情况改善,间质疏松,闭锁卵泡数量明显减少(P<0。01);与模型组比较,细胞色素P450家族基因mRNA表达水平显著提高(P<0。05),凋亡相关蛋白mRNA表达水平显著降低(P<0。05)。结论 补肾还精方通过提高细胞色素P450家族基因的表达,抑制衰老凋亡基因的表达,缓解环磷酰胺诱发的卵巢颗粒细胞损伤及闭锁卵泡的产生,实现对卵巢早衰小鼠的治疗作用。
Mechanism of Bushen Huanjing Formula(补肾还精方)Promoting Expressions of Cytochrome P450 Genes to Improve Premature Ovarian Failure
Objective To explore the molecular mechanism of Bushen Huanjing Formula(补肾还精方)in improving the pre-mature ovarian failure caused by cyclophosphamide in mice.Method Sixty 10-week-old female C57/BL6 mice were randomly divided into five groups,with 12 mice in each group,namely the blank group,the model group,the Bushen Huanjing Formula low-dose group,the Bushen Huanjing Formula medium-dose group and the Bushen Huanjing Formula high-dose group.The mice were injected with cyclophosphamide intraperitoneally to establish a model,and were treated with different concentrations of Bushen Huanjing Formula by gavage.H&E staining was used to detect the pathological morphology of mouse ovary and count fol-licles at various types.The qPCR method was used to detect mRNA expression levels of cytochrome P450 family related proteins and cell senescent apoptotic protein genes.Results Compared with that in the model group,the ovarian atrophy in mice treated with drugs improved,with loose interstitial tissue and significantly reduced atretic follicles(P<0.01).Compared with those in the model group,the mRNA expression levels of cytochrome P450 family genes were significantly increased(P<0.05),while the mRNA expression levels of apoptosis-related proteins were significantly decreased(P<0.05).Conclusion By enhancing the ex-pressions of P450 family-related proteins and inhibiting the expressions of senescence-associated apoptosis proteins,Bushen Huanjing Formula alleviates the injury of ovarian granulosa cells induced by cyclophosphamide and the production of atretic folli-cles to realize the therapeutic effect on premature ovarian failure mice.
