Mechanism of Guipi Decoction(归脾汤)Improving Chemotherapy Myasthenia Based on Network Pharmacology and Bioinformatics
Objective The effect of Guipi Decoction(归脾汤)on chemotherapy myasthenia was analyzed by network pharma-cology and bioinformatics and the possible mechanism was elucidated.Methods The potential targets of Guipi Decoction were searched through TCM system Pharmacology database and Analysis Platform(TCMSP),the related targets of myasthenia were searched through gene expression comprehensive database(GEO),and the common targets of Guipi Decoction and myasthenia were obtained by Venn software.Cytoscape software was used to select and obtain the network diagram of active ingredients-tar-get-related diseases of Guipi Decoction,and MCODE plug-in was used to select and obtain the key targets of Guipi Decoction to improve myasthenia.Single sample gene set enrichment analysis(ssGSEA)was used to explore the core immune cells that Guipi Decoction might be involved in improving myasthenia.Function and pathway enrichment analysis was conducted by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG),and the network diagram of"Guipi Decoction-key tar-get-major pathway-myasthenia"was constructed.Results A total of 394 drug-targeting genes and 914 targets related to myas-thenia were obtained.MCODE network was constructed to obtain 35 keycluster genes in which Guipi Decoction improved myas-thenia.The ssGSEA result showed that chemokine receptors and TNF family member receptors had the highest correlation with keycluster genes.According to GO enrichment analysis,the biological process of Guipi Decoction to improve myasnia mainly in-volved the regulation of gene expression,cell biosynthesis and macromolecular metabolism,etc.KEGG enrichment analysis showed that it mainly involved coronavirus disease,Chagas disease,IL-17 signaling pathway,cancer,toll-like receptor signaling pathway and TNF signaling pathway.Conclusion In summary,Guipi Decoction may inhibit intramusculature inflammation through multi-target and multi-pathway,and induce the activation of HIF-1 α to combat the oxidative stress caused by inflammation,so as to alleviate chemotherapy muscle weakness.
