基于TGF-β1/Smad3信号通路探讨甘露消毒汤对SARS-CoV-2假病毒感染hACE2转基因小鼠的调控机制

目的 利用SARS-CoV-2假病毒技术,通过TGF-β1/Smad3信号通路相关蛋白的表达变化,探讨GXT对SARS-CoV-2感染的作用机制。方法 将60只实验小鼠,随机分为正常组、模型组、甘露消毒汤组,除正常组小鼠外,其余2组小鼠运用SARS-CoV-2假病毒滴鼻进行造模,造模后进行药物干预,于药物干预的第7天进行取材。测定各组小鼠的各类指标、肺组织病理形态改变,同时运用ELISA法测定IL-10、TNF-α、IL-1β、IL-6的表达;运用qPCR法测定TGF-β1、Smad3、CD9、CD63的mRNA含量;采用WB法检测TGF-β1、Smad3、CD9、CD63的蛋白表达。结果 模型组:小鼠中的肺指数以及肺组织病理评分升高,在HE染色后观察到小鼠的肺泡间隔增厚并伴有炎性细胞浸润,组织中TGF-β1、Smad3、CD9、CD63mRNA及蛋白的表达升高,血清中TNF-α、IL-1β、IL-6含量升高,IL-10表达下降(P<0。05);GXT组:肺指数、肺组织病理评分下调(P<0。05),HE染色可见炎性细胞浸润减少,小鼠肺组织中TGF-β1、Smad3、CD9、CD63mRNA及蛋白表达降低(P<0。05),血清中TNF-α、IL-1 β、IL-6含量降低,IL-10含量升高(P<0。05)。结论 GXT能够通过下调炎性因子IL-6、TNF-α、IL-1β,上调抗炎因子IL-10,抑制炎性病理反应过程,减轻假病毒小鼠肺组织炎性反应,其机制可能与抑制TGF-β1/Smad3通路有关。
Exploring Regulatory Mechanism of Ganlu Xiaodu Decoction(甘露消毒汤)on SARS-CoV-2 Pseudovirus Infected hACE2 Transgenic Mice Based on Signal Pathway TGF-β1/Smad3
Objective Using SARS-CoV-2 pseudovirus technology,the expression changes of TGF-β1/Smad3 signaling pathway related proteins were investigated to explore the mechanism of Ganlu Xiaodu Decoction(甘露消毒汤)on SARS-CoV-2 infection.Methods Sixty experimental mice were randomly assigned to three groups:normal group,model group,Ganlu Xiaodu Decoction group.Except the normal group,the mice in the other two groups were given nasal drops of SARS-CoV-2 pseudovir-us for modeling.Following modeling,the drug intervention was administered,and the samples were collected on the seventh day of treatment.The expressions of interleukin-10(IL-10),tumor necrosis factor-α(TNF-α),interleukin 1 β(IL-1 β)and inter-leukin-6(IL-6)in serum were determined using ELISA method.The mRNA contents of transforming growth factor-β1(TGF-β1),Smad3,CD9 and CD63 in lung tissue were quantified using qPCR method.The protein expressions of TGF-β1,Smad3,CD9 and CD63 were analyzed by Western blot(WB)assay.Results Model group:The lung index and pathological score of lung tissue in mice were increased.After HE staining,HE staining revealed thickening of alveolar septa,infiltration of inflammatory cells and increased expressions of TGF-β1,Smad3,CD9 and CD63 mRNA and protein in lung tissue.Results Serum levels of TNF-α,IL-1 β and IL-6 were elevated,while IL-10 expression was decreased(P<0.05).Ganlu Xiaodu Decoction group:Lung index and lung tissue pathological score were significantly down-regulated(P<0.05).HE staining demonstrated reduced inflammatory cell infiltration and decreased expressions of TGF-β1,Smad3,CD9 and CD63 mRNA and protein in mouse lung tissue(P<0.05).HE staining showed reduced inflammatory cell infiltration,decreased expressions of TGF-β1,Smad3,CD9 and CD63mRNA and protein in mouse lung tissue(P<0.05),decreased serum levels of TNF-α,IL-1 β and IL-6 and in-creased IL-10 content(P<0.05).Conclusion Ganlu Xiaodu Decoction can down-regulate inflammatory factors(IL-6,TNF-α,IL-1 β),up-regulate anti-inflammatory factor(IL-10),inhibit the process of inflammatory pathological reactions and alleviate the inflammatory response of lung tissue in mice with pseudovirus.Its mechanism may be related to the inhibition of pathway TGF-β1/Smad3.
