Experimental Study on the Treatment of Melasma Model Mice by Carthamus Tinctorius Poria Cocos Panax Notoginseng and its Composition
Objective To observe the therapeutic effect and acute toxicity of extracts of safflower,poria cocos,Panax notogin-seng and their compositions on melasma model mice.Methods According to the literature,progesterone combined with UV irradi-ation was used to establish melasma model mice,a total of 100 Kunming mice were divided into 10 groups:blank control group,model control group,Safflower extract high-dose and low-dose group,Poria cocos high-dose and low-dose group,Panax notoginseng high-dose and low-dose group,composition high-dose and low-dose group.After 30 days of continuous admin-istration,the contents of melanocytes and lipofuscin(LF),superoxide dismutase(SOD),malondialdehyde(MDA),tyrosinase(TYR)and lipofuscin(LF)in liver tissue and back skin tissue of mice were detected,at the same time,immunohistochemical method was used to observe the changes of skin melanocytes of mice after administration.Finally,the acute toxicity test of mix-tures of safflower,poria and notoginseng in mice was carried out with the maximum dose experiment.Results Compared with model group,safflower extract,Poria cocos extract,Panax notoginseng extract and their compositions could increase SOD activity and decrease MDA,TYR and LF contents in liver and skin of mice,the number of melanocytes in the skin tissue of model mice was also reduced,the differences were statistically significant(P<0.01,P<0.05);The acute toxicity test of mice showed no death in each group and normal activity performance.Compared with the control group,the body weight of mice had no signifi-cant difference(P>0.05).Conclusion The effect of safflower,poria cocos,Panax notoginseng and their compositions on im-proving melasma was related to the increase of antioxidant capacity and the decrease of tyrosinase content in mice,and no obvious acute toxicity was observed in animals under the experimental conditions.
