光敏汤对HaCaT细胞表达IL-10影响研究

摘要

目的:研究光敏汤对角质形成细胞HaCaT细胞表达IL-10的影响,初步探讨光敏汤的光保护作用.方法:将HaCaT细胞分为光敏汤Ⅰ号组、光敏汤Ⅱ号组、甲氨蝶呤(MTX)阳性对照组、空白对照组、UVB对照组.HaCaT细胞以30mJ/cm2剂量的UVB照射后培养6h、12h、24h、48h采用ELISA法检测各组HaCaT细胞IL-10水平.结果:UVB辐射可增强HaCaT细胞IL-10表达;UVB照射后48h光敏汤Ⅰ号组、光敏汤Ⅱ号组、甲氨蝶呤(MTX)阳性对照组均可有效降低HaCaT细胞分泌IL-10,组间比较差异有统计学意义(P<0.05);UVB照射后72h光敏汤Ⅱ号组的效果优于甲氨蝶呤(MTX)阳性对照组,组间比较差异有统计学意义(P<0.05);光敏汤Ⅰ号组与甲氨蝶呤(MTX)阳性对照组组间比较差异无统计学意义(P>0.05).结论:光敏汤Ⅱ号预防UVB照射所致炎症比传统药物效果更佳,能减少产生HaCaT细胞IL-10.

Abstract

Objective:To observe the effect of Photosensitive Decoction on expression of IL-10 in the keratinocyte HaCaT cells and preliminary study on the photoprotection of Photosensitive Decoction. Method: HaCaT cells were divided into 5 groups, Photosensitive DecoctionⅠgroup, Photosensitive Decoction Ⅱ group, MTX positive control group, blank control group and UVB control group. HaCaT cells were cultured for 6 h, 12 h, 24 h and 48 h after the UVB irradiation with the dosage 30mJ/cm2. The level of IL-10 in HaCaT cells of each group was detected by ELISA method. Result: UVB irradiation could increase the expression of IL-10 in HaCaT cells. 48 h after the UVB irradiation, Photosensitive DecoctionⅠgroup, Photosensitive Decoction Ⅱ group and MTX positive control group could decrease the secretion of IL-10 in HaCaT cells effectively. There was statistical significance compared among groups(P<0.05). 72 h after the UVB irradiation, the effect of Photosensitive Decoction Ⅱ group was superior to that of MTX positive control group. There was statistical significance compared with Photosensitive DecoctionⅡgroup and MTX positive control group(P<0.05). There was no statistical significance compared with Photosensitive DecoctionⅠgroup and MTX positive control group(P>0.05). Conclusion: Photosensitive Decoction Ⅱ has better effect in preventing inflammation due to UVB irradiation than traditional medicine, which could decrease the production of IL-10 in HaCaT cells effectively.
