不同提取方式对三七改善小鼠睡眠功能的影响

目的:观察不同提取方式对三七改善小鼠睡眠功能的影响。方法:通过直接睡眠试验、延长戊巴比妥钠睡眠时间试验、戊巴比妥钠阈下剂量催眠试验、巴比妥钠睡眠潜伏期试验,评价三七水提物、三七30%乙醇提取物、三七70%乙醇提取物对小鼠睡眠的影响。结果:与对照组比较,三七70%乙醇提取物各剂量组、三七30%乙醇提取物高剂量组、三七水提物高剂量组均可延长戊巴比妥钠诱导的小鼠睡眠时间(P<0。05或P<0。01),增加戊巴比妥钠阈下催眠剂量入睡小鼠只数(P<0。05或P<0。001),缩短小鼠的睡眠潜伏期(P<0。05或P<0。001)。以小鼠睡眠时间、入睡只数、睡眠潜伏期时间作为评估不同提取物对改善小鼠睡眠作用的主要指标。三七30%乙醇提取物低剂量组、三七水提物低剂量组与对照组比较改善小鼠睡眠作用不显著,与三七水提取物高剂量组比较三七70%乙醇提取物高剂量组对小鼠睡眠改善作用差异显著(P<0。05),三七30%乙醇提取物高剂量组以及三七70%乙醇提取物低剂量组改善小鼠睡眠作用无显著性差异(P>0。05)。三七70%乙醇提取物高剂量组、三七30%乙醇提取物高剂量组、三七水提物高剂量组小鼠下丘脑及血清中的5-羟色胺(5-HT)及γ-氨基丁酸(GABA)含量均显著高于对照组(P<0。05或P<0。01),多巴胺(DA)含量均显著低于对照组(P<0。05或P<0。01)。结论:三七水提物、三七30%乙醇提取物、三七70%乙醇提取物均可在一定程度上改善小鼠的睡眠,其中三七70%乙醇提取物效果最佳。
Objective:To observe the effect of different extraction methods on the improvement of sleep function in mice by Panax notoginseng.Methods:The effects of water extract,30%ethanol extract and 70%ethanol extract of Panax notoginseng on sleep in mice were evaluated by direct sleep test,prolonged pentobarbital sodium sleep time test,pentobarbital sodium subthreshold dose hypnosis test and barbital sodium sleep latency test.Results:Compared with the control group,each dose group of 70%ethanol extract of Panax notoginseng,the high dose group of 30%ethanol extract of Panax notoginseng and the high dose group of water extract of Panax notoginseng could prolong the sleep time of mice induced by pentobarbital sodium(P<0.05 or P<0.01),increase the number of mice falling asleep at the subliminal hypnotic dose of pentobarbital sodium(P<0.05 or P<0.001)and shorten the sleep latency of mice(P<0.05 or P<0.001).The sleep time,the number of sleeping mice and the sleep latency time were used as the main indicators to evaluate the effect of different extracts on improving sleep in mice.Compared with the control group,the low-dose group of 30%ethanol extract of Panax notoginseng and the low-dose group of water extract of Panax notoginseng had no significant effect on improving sleep in mice.Compared with the high-dose group of water extract of Panax notoginseng,the high-dose group of 70%ethanol extract of Panax notoginseng had significant difference in improving sleep in mice(P<0.05).The contents of 5-hydroxytryptamine(5-HT)and γ-aminobutyric acid(GABA)in hypothalamus and serum of mice in the high dose of 70%ethanol extract group,the high dose of 30%ethanol extract group and the high dose of water extract group were significantly higher than those in control group(P<0.05 or P<0.01),and the content of dopamine(DA)was significantly lower than that in control group(P<0.05 or P<0.01).Conclusion:Panax notoginseng water extract,30%ethanol extract of Panax notoginseng and 70%ethanol extract of Panax notoginseng could improve the sleep of mice to a certain extent,among which 70%ethanol extract of Panax notoginseng has the best effect.
