板蓝根对肺炎支原体小鼠模型细胞因子IL-2 IL-4TNF-α的影响

目的:通过建立肺炎支原体感染小鼠模型,观察不同浓度下的板蓝根提取物对肺炎支原体小鼠的细胞因子IL-2,IL-4,TNF-α的影响。方法:50只SPF级BALB/c健康小鼠随机分成对照组、模型组、板蓝根高浓度组(250mg/mL),中浓度组(125mg/mL),低浓度组(62。5mg/mL),每组10只。模型组用滴鼻法建立肺炎支原体感染模型,对照组小鼠滴入等量的PBS液。对建模成功的小鼠进行灌胃治疗7天,建立板蓝根不同浓度组,提取肺组织。用苏木精-伊红(HE)染色法研究小鼠的肺组织病理变化。用流式微球阵列法(CBD)检测血清炎症因子IL-2、IL-4和TNF-α的表达水平。结果:与对照组比较模型组小鼠肺湿重指数升高(P<0。05),与模型组比较板蓝根高中低浓度组的肺湿重指数、病理评分降低(P<0。05),与对照组比较模型组IL-2、IL-4和TNF-α浓度升高(P<0。05),与模型组比较板蓝根高浓度组,中浓度组,低浓度组IL-2浓度降低(P<0。05),板蓝根的低浓度组IL-4浓度降低(P<0。05),板蓝根高浓度组TNF-α浓度降低(P<0。05)。结论:肺炎支原体感染小鼠引起肺病理生理改变,细胞因子IL-2,IL-4,TNF-α释放,板蓝根不同浓度能不同程度地抑制细胞释放,对肺组织起到保护作用。
Objective:To observe the effects of different concentrations of Radix Isatidis extract on cytokines IL-2,IL-4 and TNF-α in mycoplasma pneumoniae mice by establishing a mouse model of mycoplasma pneumoniae infection.Methods:Fifty SPF BALB/c healthy mice were randomly divided into control group,model group,high concentration of Radix Isatidis group(250mg/mL),medium concentration group(125mg/mL),low concentration group(62.5mg/mL),with 10 mice in each group.The mycoplasma pneumoniae infection model was established by nasal drip in the model group,and the mice in the control group were dripped with the same amount of PBS solution.The mice with successful modeling were treated with gavage for 7 days,and different concentration groups of Radix Isatidis were established to extract lung tissue.The pathological changes of lung tissue in mice were studied by hematoxylin-eosin(HE)staining.The expression levels of serum inflammatory factors IL-2,IL-4 and TNF-α were detected by flow microsphere array(CBD).Results:Compared with the control group,the lung wet weight index of the model group increased(P<0.05).Compared with the model group,the lung wet weight index and pathological score of the high,medium and low concentration groups of Radix Isatidis decreased(P<0.05).Compared with the control group,the concentration of IL-2,IL-4 and TNF-α in the model group increased(P<0.05).Compared with the model group,the concentration of IL-2 in the high concentration group,medium concentration group and low concentration group of Radix Isatidis decreased(P<0.05).Compared with the model group,the concentration of IL-4 in the low concentration group of Radix Isatidis decreased(P<0.05),and the concentration of TNF-α in the high concentration group of Radix Isatidis decreased(P<0.05).Conclusion:Mycoplasma pneumoniae infection in mice caused lung pathophysiological changes,cytokine IL-2,IL-4,TNF-α release,different concentrations of Radix Isatidis can inhibit cell release to varying degrees,play a protective role in lung tissue.
