Effect and mechanism of Polydatin on hyperlipidemic mice by inhibiting oxidative stress and inflammation
Objective To explore the effect and mechanism of polydatin on oxidative stress and inflammation in hyperlipidemic mice.Methods Hyperlipidemia mouse model were established on high-fat chow and randomly divided into model group,polyda-tin group,simvastatin group,and another normal group,ten mice in each group.The polydatin and simvastatin groups were given corresponding drugs by gavage,the normal and model groups were replaced with equal volume normal saline.After 28 days,The body mass and liver index were measured in each group of mice;Total cholesterol(TC),triglyceride(TG),low density lipopro-tein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were measured using a fully automated biochem-ical analyzer;Pathological changes in the liver were examined by hematoxylin-eosin(HE)staining and oil red O(ORO)stai-ning;ELISA for ultrasensitive C-reactive protein(hs-CRP),interleukin-1 β(IL-1 β)and interleukin-6(IL-6)expres-sion levels in serum;Detection of nuclear factor-κB p65(NF-κB p65)and tumour necrosis factor-α(TNF-α)mRNA ex-pression in aortic tissues by qPCR;The protein expressions of tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)and nuclear factor-κB p65(NF-κB p65)in aorta of mice were determined by Western Blot;The activities of malondialdehyde(MDA),glutathione peroxidase(GSH)and superoxide dismutase(SOD)in liver tissues of mice were measured by thiobarbitu-ric acid,dithiobarbituric acid and water-soluble tetrazolium methods,respectively.Results Compared with the normal group,the body mass and liver index of mice in the model group were significantly increased(P<0.01);TC,TG and LDL-C levels in serum were significantly elevated and HDL-C was significantly decreased(P<0.01),and liver tissues showed significant steatosis;the levels of inflammatory factors in serum,such as hs-CRP,IL-1β and IL-6 were significantly increased(P<0.01);the expressions of TNF-α and NF-κB p65 mRNA is significantly elevated in mouse aorta(P<0.01);the protein expressions of IFN-γ,TNF-α and NF-κB p65 in mouse aorta were increased(P<0.01);MDA levels were significantly increased(P<0.01)and GSH and SOD levels were significantly decreased(P<0.01)in liver tissues.Compared with the model group,the body mass of mice in the polydatin and simvastatin groups were not statistically dif-ferent(P>0.05),but the trend of body weight gain became slower and the liver index was significantly lower(P<0.05).After treatment with polydatin,TC,TG and LDL-C in serum were significantly reduced(P<0.01),HDL-C was significantly in-creased(P<0.01),and histopathology showed significant improvement in liver steatosis;The hs-CRP,IL-1β and IL-6 lev-els in serum were significantly reduced(P<0.01);The mRNA and protein expressions of TNF-α and NF-κB p65 in aortic tissues were significantly reduced(P<0.05,P<0.01).The level of MDA was significantly reduced(p<0.01),and the levels of GSH and SOD were significantly increased in liver(P<0.05,P<0.01).Conclusion Polydatin has a certain protective effect on hyperlipidemia mice,The mechanism may be related to its ability to reduce inflammatory responses and alleviate oxidative stress damage.
