补肾活血方对复发性流产患者血清VEGF、HIF-1α、TGF-β1、sflt-1影响研究

摘要
目的 研究补肾活血方对APL阳性RSA患者外周血VEGF、HIF-1α、TGF-β1、sflt-1、miR-423-5p蛋白表达量影响.方法 选取APL阳性RSA患者60例,分别于孕前给予药物调理.其中观察组服用补肾活血方,对照组服用肠溶阿司匹林,均治疗3个月.两组治疗前后分别检测血清VEGF、HIF-1α、TGF-β1、sflt-1、miR-423-5p蛋白表达及中医证候积分.结果 两组治疗后VEGF蛋白表达均显著升高;与对照组比较,观察组VEGF蛋白表达量显著高于对照组;两组治疗前后,HIF-1α蛋白表达量均无显著性差异;治疗后两组TGF-β1蛋白表达显著升高,与对照组比较,观察组TGF-β1蛋白表达显著升高;治疗后两组sflt-1蛋白表达显著降低;与对照组比较,观察组sflt-1蛋白表达显著降低.两组治疗后miR-423-5p相对表达量均降低,观察组优于对照组(P＜0.05).观察组中医证候疗效比较,治疗组优于对照组(P＜0.05).结论 补肾活血方应用于RSA患者孕前调理,可以通过上调VEGF、TGF-β1蛋白表达,下调sflt-1、miR-423-5p蛋白表达等途径,改善APL对子宫内膜的不良影响;同时可有效降低中医证候积分,从而降低再次妊娠后流产的发生率.
Abstract
Objective To study the effect of Bushen Huoxue Formula on peripheral blood VEGF,HIF-1α,TGF in APL positive RSA patients-β1.The expression levels of sflt-1 and miR-423-5p proteins are affected.Methods Sixty APL positive RSA patients were selected and administered medication before pregnancy.The observation group received the tonifying kidney and ac-tivating blood formula,while the control group received enteric coated aspirin,both receiving treatment for 3 months.Serum VEGF,HIF-1α,and TGF were measured before and after treatment in both groups-β1.Expression of sflt-1 and miR-423-5p proteins and integration of traditional Chinese medicine syndromes.Results The expression of VEGF protein was significantly increased in both groups after treatment;Compared with the control group,the expression of VEGF protein in the observation group was significantly higher than that in the control group;HIF-1 before and after treatment in both groups α There was no sig-nificant difference in protein expression levels;Two groups of TGF after treatment-β1.The protein expression was significantly increased,and compared with the control group,the observation group had TGF-β1.The protein expression significantly in-creased;After treatment,the expression of sflt-1 protein significantly decreased in both groups;Compared with the control group,the expression of sflt-1 protein in the observation group was significantly reduced.After treatment,the relative expression of miR-423-5p decreased in both groups,and the observation group was better than the control group(P＜0.05).The obser-vation group compared the efficacy of traditional Chinese medicine syndromes,and the treatment group was better than the control group(P＜0.05).Conclusion Bushen Huoxue Recipe can be used to regulate RSA patients before pregnancy by up regulating VEGF and TGF-β1 protein expression,downregulation of sflt-1,miR-423-5p protein expression,and other pathways to im-prove the adverse effects of APL on the endometrium;At the same time,it can effectively reduce the traditional Chinese medicine syndrome score and reduce the incidence of miscarriage after another pregnancy.
