高效液相色谱法同时测定还脑益聪方中5种川芎成分含量

摘要
目的 建立还脑益聪方中阿魏酸、洋川芎内酯Ⅰ、洋川芎内酯A、藁苯内酯和欧当归内酯A 5种成分含量测定方法,探究其物质基础.方法 采用高效液相色谱法,色谱柱Agilent Extend-C18(4.6 mm ×250 mm,5pm);流动相:甲醇(A),0.3%磷酸溶液(B),乙腈(C);流速:1m L/min;柱温:30℃;检测波长:280nm.结果 还脑益聪方中川芎的5种化学成分均被检测到,建立的方法无阴性干扰,5种成分均达到良好的分离效果,标准曲线线性范围良好.结论 该方法简便稳定,重复性好,可用于还脑益聪方中川芎成分的定量检测,为还脑益聪方治疗AD的物质基础提供依据.
Abstract
Objective To establish a method for the determination of 5 components:Ferulic acid,Ligusticolide Ⅰ,Ligusticolide A,Ligustilide and Angelica lactone A in Huannao Yicong Decoction,and explore the material basis.Methods HPLC was per-formed on Agilent Extend-C18(4.6 mm ×250 mm,5μm)column.Mobile phase:methanol(A),0.3%phosphoric acid solu-tion(B),acetonitrile(C);Flow rate:1m L/min;Column temperature:30℃;Detection wavelength:280nm.Results A total of 5 chemical constituents of chuanxiong were detected in Huannao Yicong Decoction.The established method has no negative inter-ference,and the five components achieved good separation effect,and the linear range of standard curve is good.Conclusion This method is simple,stable and reproducible,and can be used for the quantitative detection of Chuanxiong in Huannao Yicong prescription,providing evidence for the material basis in the treatment of AD.
