电针预处理对异丙肾上腺素诱发小鼠急性心肌缺血的保护作用研究

目的 探讨电针预处理"额旁1线"对异丙肾上腺素(ISO)诱发小鼠急性心肌缺血的保护效应及可能机制。方法 30只雄性昆明小鼠随机分为对照组、模型组和电针预处理组,每组10只。模型组和电针预处理组通过背部皮下多点位注射ISO诱发小鼠急性心肌缺血模型,对照组皮下注射生理盐水。电针预处理组电针小鼠头穴双侧"额旁1线"区域,疏密波,频率2 Hz/100 Hz,强度2mA,每次电针30min,每天1次,连续7天,第6天和第7天电针后注射ISO造模。末次给药造模2h后,对小鼠进行心电图检测;取心肌组织经苏木素-伊红(HE)和masson染色观察小鼠心肌形态学变化,透射电镜观察心肌细胞超微结构变化;采用酶联免疫吸附试验(ELISA)测定小鼠心肌组织白细胞介素-6(IL-6),肿瘤坏死因子-α(TNF-α)及血清肌酸激酶同工酶MB(CK-MB)、肌钙蛋白cTnⅠ含量。结果 模型组小鼠造模后心电图T波高度降低、J点位移明显(P<0。01),电针预处理组小鼠造模后心电图T波高度、J点电压减小不显著,与对照组相比,模型组小鼠心肌TNF-α、IL-6以及血清CK-MB、cTnⅠ均显著增高(P<0。01),形态学观察显示缺血心肌组织出现明显损伤。与模型组相比,电针预处理组小鼠血清,CK-MB、cTn Ⅰ及血清心肌TNF-α、IL-6水平明显降低(P<0。01),形态学观察缺血心肌损伤程度显著减轻。结论 电针预处理"额旁1线"能明显改善急性心肌缺血小鼠心电图T波及J点偏移,减轻心肌病理损伤程度,降低心肌CK-MB、cTnⅠ及血清IL-6、TNF-α含量等发挥心肌保护作用。
Study on the myocardial protective effect of electroacupuncture pretreatment on isoprot-erenol induced acute myocardial ischemia in mice
Objective To explore the protective effect and possible mechanism of electroacupuncture pretreatment on isoproterenol(ISO)induced acute myocardial ischemia in mice.Methods 30 male Kunming mice were randomly divided into a control group,a model group,and an electroacupuncture pretreatment group,with 10 mice in each group.The model group and electroacupunc-ture pretreatment group induced acute myocardial ischemia model in mice by subcutaneous injection of ISO at multiple points on the back,while the control group received subcutaneous injection of physiological saline.The electroacupuncture pretreatment group treated mice with electroacupuncture in the bilateral"Later line 1 of forehead"area of the head point,with a density wave,frequency of 2 Hz/100 Hz,intensity of 2mA,and electroacupuncture for 30 minutes each time,once a day,for 7 consecutive days.After electroacupuncture on the 6th and 7th days,ISO models were injected.Two hours after the last administration of the drug for modeling,the mice were subjected to electrocardiogram testing;The myocardial tissue of mice was stained with hematoxy-lin eosin(HE)and masson to observe the morphological changes of myocardium,and the ultrastructural changes of myocardial cells were observed under transmission electron microscope;Enzyme linked immunosorbent assay(ELISA)was used to determine the levels of interleukin-6,tumor necrosis factor-α in mouse myocardial tissue,and creatin kinase-MB,cardiac troponin Ⅰ in serum.Results After modeling,the height of ECG T-wave and the displacement of J-point were significantly decreased in model group(P<0.01),but the height of ECG T-wave and the voltage of J-point were not significantly decreased in electroa-cupuncture pretreatment group.Compared with the control group,the myocardial TNF-α,IL-6,serum CK-MB and cTnⅠ in model group were significantly increased(P<0.01),and morphological observation showed that ischemic myocardial tissue was obviously damaged.Compared with the model group,the serum CK-MB,cTnⅠ,myocardial TNF-α and IL-6 levels in the electroacupuncture preconditioning group were significantly decreased(P<0.01),and the degree of ischemic myocardial injury was significantly reduced by morphological observation.Conclusion Electroacupuncture pretreatment of"Later line 1 of forehead"can obviously improve the deviation of ECG T wave and J point in mice with acute myocardial ischemia,reduce the degree of myo-cardial pathological injury,and reduce the contents of CK-MB,cTnⅠ,IL-6 and TNF-α in myocardium,thus playing a pro-tective role.
