黄芩汤调控TLR2/NF-κB/NLRP3通路对溃疡性结肠炎细胞焦亡的影响

目的 基于TLR2/NF-KB/NLRP3通路探讨黄芩汤对溃疡性结肠炎细胞焦亡的影响.方法 实验分为空白组、模型组、低中高中药血清组.LPS+ATP构建焦亡模型,cck-8法检测生长情况,流式细胞术检测凋亡,Elisa检测IL-18、IL-1β、TNF-α 水平,qRT-PCR 检测 IL-1β、NLRP3、Caspase-1、IL-18、ASC、GSDMD mRNA 表达情况,WB 检测 TLR2、p-NF-κB-p65、GSDMD蛋白水平.结果 与模型组相比,三组中药血清细胞活力均明显上升(P<0.05)、凋亡率均显著降低(P<0.01).与模型组相比,三组中药血清IL-18、IL-1β、TNF-α水平均明显降低(P<0.05).与模型组相比,中剂量组 Caspase-1、NLRP3、ASC、IL-18 的 mRNA 表达明显降低(P<0.05),高剂量组 IL-1β、NLRP3、Caspase-1、IL-18、ASC、GSDMD的mRNA表达均显著降低(P<0.01).与模型组相比,中、高剂量组TLR2、p-NF-κB-p65及高剂量组GS-DMD蛋白表达显著降低(P<0.01).结论 黄芩汤抑制溃疡性结肠炎焦亡的机制可能是调控TLR2/NF-κB/NLRP3通路.
Effect of Huangqin Decoction regulating TLR2/NF-κB/NLRP3 pathway on pyroptosis of ulcerative colitis
Objective To investigate the regulatory effect of Huangqin Decoction on the pyroptosis pathway of TLR2/NF-κB/NLRP3/in ulcerative colitis.Methods HCT-116 cells were randomly divided into blank group,model group,low,medium and high herbal serum group(15%,20%and 25%herbal serum)LPS+ATP was used to construct the pyroptosis model,Cell growth was detected by cck-8 method,cell apoptosis was detected by flow cytometry,IL-18,IL-1β and TNF-α levels were detec-ted by Elisa,mRNA expression of IL-1β,NLRP3,Caspase-1,IL-18,ASC and GSDMD were detected by RT-PCR.The protein levels of TLR2,p-NF-κB-p65and GSDMD were detected by WB.Results Compared with the model group,the ser-um cell vitality of the three groups was obviously increased(P<0.05),and the apoptosis rate was significantly decreased(P<0.01).Compared with model group,serum levels of IL-18,IL-1β and TNF-α in three groups were obviously decreased(P<0.05).Compared with model group,mRNA expressions of Caspase-1,NLRP3,ASC and IL-18 in medium-dose group were obviously decreased(P<0.05),while mRNA expressions of IL-1β,NLRP3,Caspase-1,IL-18,ASC and GSDMD in high-dose group were significantly decreased(P<0.01).Compared with model group,the protein expressions of TLR2,P-NF-Kb-p65 and GSDMD in medium and high dose groups were significantly decreased(P<0.01).Conclusion The mecha-nism of Huangqin Decoction inhibiting pyrodeath in ulcerative colitis may be the regulation of TLR2/NF-κB/NLRP3 pathway.
