Effect of acupuncture on the expression of miR-29a-3p and miR-29b-3p in hipp-ocampal tissue of rats with cerebral ischemia-reperfusion injury
Objective To observe the expression of miR-29a-3p and miR-29b-3p in the hippocampal region of the ischemic side of rats with cerebral ischemia-reperfusion injury(CIRI)after 72 h of acupuncture intervention,and to explore the mecha-nism of acupuncture to promote anti-CIRI repair.Methods Thirty SD rats were randomized to sham-operated group,model group and acupuncture group,10 rats in each group,the modeling group was prepared using the thread bolus method,the sham-operated group only stripped the blood vessels without inserting the thread bolus;the sham-operated group and model group were only tied without acupuncture,the acupuncture group was tied and then acupuncture was applied to"Dazhui","Baiji"and"Shuigu".In the sham-operated group and the model group,only the vessels were stripped and no needles were inserted.Garcia's score was used to assess neurological function,TTC staining was used to detect the ratio of cerebral infarct area,and qRT-PCR was used to detect the expression of miR-29a-3p and miR-29b-3p in the hippocampus on the ischemic side.Results Com-pared with the sham-operated group,the Garcia score was significantly lower(P<0.01),the cerebral infarct area ratio was sig-nificantly increased(P<0.01),and the expression of miR-29a-3p and miR-29b-3p in the hippocampal region on the af-fected side was significantly lower(P<0.01)in the model and needle groups;compared with the model group,the Garcia score was significantly higher(P<0.01)in the needle group,the cerebral infarct area ratio was significantly decreased(P<0.01),and the expression of miR-29a-3p and miR-29b-3p in the affected hippocampal tissue was significantly increased(P<0.01).Conclusion Acupuncture can reduce the area of cerebral infarction on the ischemic side,improve neurological impairment and exert cerebral protective effects in CIRI rats,which may be related to the upregulation of miR-29a-3p and miR-29b-3p expression in the hippocampal region on the ischemic side after 72 h of intervention.
