葛粉对小鼠糖脂代谢紊乱的改善作用研究

目的 探究长期摄入葛粉对糖脂代谢紊乱的改善作用。方法 将36只C57BL/6J小鼠随机分为空白组、对照组和葛粉组,每组12只。基于高脂饮食联合葡聚糖硫酸钠诱导的小鼠糖脂代谢紊乱模型。葛粉组灌胃5g/kg体重的葛粉,其余两组灌胃等量生理盐水。每周测量各组小鼠体重并进行疾病活动度评分,15周后检测小鼠空腹血糖和葡萄糖耐性,对小鼠结肠长度、肝脏及脂肪重量进行测量并通过试剂盒检测小鼠血清中甘油三酯(TG)、胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)水平。综合以上实验结果评估葛粉对小鼠糖脂代谢紊乱的改善作用。结果 高脂饮食联合葡聚糖硫酸钠诱导后小鼠体重出现非正常快速增长,葛粉饮食干预后增长速度降低。与空白组相比,对照组小鼠疾病活动度评分显著升高,结肠缩短且伴随肝脏及脂肪组织重量的显著增加;葛粉饮食干预后,各相关指标均有显著改善,表明长期摄入葛粉能够缓解小鼠肠道炎症,抑制小鼠体重非正常增长,改善肝脏及脂肪组织脂质代谢;与空白组比较,对照组小鼠空腹血糖、葡萄糖耐量、血清TG、TC、LDL-C水平均有显著增高,糖耐性、HDL-C则出现显著下降;葛粉饮食干预后所有指标均有所改善,表明葛粉饮食干预能够降低小鼠血糖,提高糖耐性,改善小鼠血脂情况。结论 摄入葛粉能一定程度预防糖脂代谢紊乱,这可能与葛粉中的抗性淀粉及异黄酮类调节糖脂代谢以及改善肠道炎症有关。
Ameliorative effect of kudzu powder on the disorder of glucose and lipid metabolism in mice
Objective Explore the improvement effect of long-term intake of Pueraria on chronic inflammation and glycolipid metabolism disorder.Methods 36 C57BL/6J mice were randomly divided into blank group,control group and kudzu powder group,with 12 mice in each group.Based on the chronic inflammation and glycolipid metabolism disorder mice model which was established by feeding high-fat diet and dextran sulfate sodium.kudzu powder group was gavage with 5g/kg body weight(BW)of kudzu powder,and the other two groups were gavage with the same amount of normal saline.The weight of mice in each group was measured weekly and the disease activity index was scored.After 15 weeks,the fasting blood glucose and glucose tolerance of mice were measured.The colon length,liver and fat weight of mice were measured.The serum TG,TC,HDL-C and LDL-C levels of mice were measured by the kit.Based on the above experimental results,the improvement effect of kudzu powder on chronic inflammation and glycolipid metabolism disorder in mice was evaluated.Results The body weight of mice increased abnor-mally and rapidly after feeding high-fat diet and DSS,and the growth rate decreased after the intervention of kudzu powder diet.Compared with the blank group,the disease activity index score of the control group was significantly higher,the colon was short-ened,and the weight of liver and adipose tissue was significantly increased.Compared with the control group,all relevant indica-tors were significantly improved after the intervention of kudzu powder diet.These results showed that that kudzu powder diet in-tervention can alleviate the intestinal inflammation of mice,inhibit the abnormal weight growth of mice,and improve the lipid me-tabolism of liver and adipose tissue.Compared with the blank group,the fasting blood glucose,glucose tolerance,serum TG,TC,LDL-C levels in the control group were significantly increased,while the glucose tolerance and HDL-C levels were signifi-cantly decreased.Compared with the control group,all indicators of the intervention group were improved.These experimental re-sults showed that kudzu powder diet intervention can reduce blood glucose,improve glucose tolerance and improve blood lipid in mice.Conclusion Kudzu powder may have a potential role in preventing prevent chronic inflammation and disorder of glucose and lipid metabolism to a certain extent,which may be related to the regulation of glucose and lipid metabolism and improvement of in-testinal inflammation by resistant starch and isoflavones in Kudzu powder.
