目的 优化艾椒黄乳膏的制备工艺,观察其对银屑病小鼠皮损的改善作用。方法 以硬脂醇、凡士林、月桂醇硫酸钠、甘油用量为考察对象,以外观性状和物理稳定性为评价指标,采用L9(34)表正交实验设计,优化艾椒黄乳膏的制备工艺,并以咪喹莫特诱导的银屑病小鼠为模型,比较艾椒黄乳膏与艾椒黄洗剂对银屑病小鼠皮损面积和严重程度(psoriasis area and severity index,PASI)评分、皮肤病理变化、皮损处增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)表达和皮损处皮肤IL-17和IL-23水平的影响。结果 最佳处方为硬脂醇1。8g,凡士林2。0g,月桂醇硫酸钠0。15g,甘油0。575g。药效学实验表明,艾椒黄乳膏可使咪喹莫特诱导的银屑病小鼠皮损处皮肤变薄、炎症减轻,PCNA表达下降,皮损皮肤中IL-17和IL-23水平降低,相较于艾椒黄洗剂更优。结论 该研究筛选的艾椒黄乳膏制备工艺稳定,乳膏质地细腻,便于携带,可明显改善银屑病小鼠皮肤的皮损现象。
Study on preparation of Aijiaohuang Cream and improvement of psoriasis mice skin le-sions
Objective To optimize the preparation technology of Aijiaohuang cream and observe its effect on psoriasis mice skin lesions.Methods With the mass of stearyl alcohol,Vaseline,sodium lauryl sulfate and glycerin as the objects of investigation,and taking cream appearance and physical stability as the evaluation indexes,the L9(34)orthogonal design was used to select cream prescription.Imiquimod-induced psoriasis mice were used as the model to compare the psoriasis area and severity index(PASI)score,skin pathological changes and the expression of proliferating cell nuclear antigen(PCNA)and levels of IL-17 and IL-23 in skin lesions of psoriasis mice with Aijiaohuang cream and Aijiaohuang lotion.Results The best prescription was stearyl alcohol 1.8g,petroleum jelly 2.0g,sodium lauryl sulfate 0.15g,glycerol 0.575g.Pharmacodynamic experiments showed that Aiqianghuang cream could reduce skin thinning,inflammation,PCNA expression and IL-17 and IL-23 levels in psoriasis mice induced by imiquimod,which was better than Aiqianghuang cream.Conclusion The preparation process of the cream screened in this study is stable,the cream is fine in texture,easy to carry,and can significantly improve the skin lesions of psori-asis mice.
