益肾祛痹通络方对肾虚寒湿痹阻型CIA大鼠miR-150-5p/SOCS1轴的调控作用

目的 探究益肾祛痹通络方对肾虚寒湿痹阻型胶原诱导关节炎大鼠的治疗作用及对miR-150-5p、细胞因子信号抑制因子1(suppressor of cytokine signaling,SOCS1)的影响。方法 20只SPF级SD大鼠,随机选取6只为正常对照组,另外14只构建肾虚寒湿痹阻型胶原诱导关节炎大鼠模型,成功10只,随机分为模型组、治疗组,各5只。治疗组予益肾祛痹通络方(0。2g/ml)灌胃治疗,每日1次,共2周。模型组及对照组均予生理盐水灌胃处理。HE染色观察踝关节软骨组织病变情况;免疫组化检测踝关节中vimentin的表达水平;qPCR检测踝关节组织中的miR-150-5p、SOCS1 mRNA表达水平;ELISA检测血清中TNF-α、IL-6的表达水平。结果 与对照组比较,模型组关节炎指数明显升高(P<0。05),HE染色表明踝关节明显狭窄,软骨损伤明显,滑膜组织增生,炎症细胞浸润;踝关节中vimentin、miR-150-5p水平升高(P<0。05),SOCS1 mRNA的表达水平无明显变化(P>0。05);血清中TNF-α、IL-6的表达水平明显升高(P<0。05)。与模型组比较,治疗组关节炎指数明显降低(P<0。05),HE染色表明关节腔未见明显狭窄,滑膜组织增生现象减少,软骨表面光滑,炎症细胞浸润降低,踝关节损伤明显改善。踝关节vimentin、miR-150-5p水平明显降低(P<0。05),SOCS1mRNA水平明显升高(P<0。05);血清中TNF-α、IL-6水平明显降低(P<0。05)。结论 益肾祛痹通络方可能通过调控miR-150-5p/SOCS1通路,抑制细胞因子表达,改善关节炎症。
Study on the Regulation of Yishen Qubi Tongluo Decoction for the Treatment of Collagen-induced Arthritis Rats with Kidney Deficiency and Cold-Dampness Syndrome Based on the miR-150-5p/SOCS1 Axis
Objective To explore the therapeutic effect of Yishen Qubi Tongluo decoction on collagen-induced arthritis(CIA)rats with kidney deficiency and cold-dampness syndrome and the influence on miR-150-5p and suppressor of cytokine signa-ling 1(SOCS1).Methods Among 20 SPF SD rats,6 cases were randomly selected as control group,and 10 of the remaining 14 cases were successfully established CIA models,and divided into model group and treatment group,5 cases respectively.The treatment group was administered intragastrically with Yishen Qubi Tongluo decoction(0.2g/ml),once per day,for 2 weeks,while model group and control group with normal saline.HE staining was used to observe the pathological changes of ankle carti-lage.Immunohistochemistry was used to detect the expression of vimentin in ankle joint.The expression levels of miR-150-5p and SOCS1 mRNA in ankle joint were detected by qPCR.The serum levels of tumor necrosis factor(TNF)-α and interleukin(IL)-6 in were detected by ELISA.Results Compared with the control group,the arthritis index of the model group was signifi-cantly higher(P<0.05);obvious ankle joint stenosis,obvious cartilage damage,synovial tissue hyperplasia,accompanied by se-rious inflammatory cell infiltration were showed in HE staining results;the levels of Vimentin and miR-150-5p in ankle joint were increased(P<0.05),but there was no significant change in SOCS1 level(P>0.05);the expressions of TNF-αand IL-6 in serum were also increased(P<0.05).Compared with the model group,the arthritis index of the treatment group was signif-icantly decreased(P<0.05);HE staining results showed that there was no obvious stenosis in the joint cavity,the proliferation of synovial tissue was reduced,the cartilage surface was smooth,the infiltration degree of inflammatory cells was reduced,and the ankle joint injury was significantly improved;the levels of vimentin and miR-150-5p in ankle joint were decreased(P<0.05),while the levels of SOCS1 were increased(P<0.05);the serum level of TNF-αand IL-6 were decreased(P<0.05).Con-clusion The Yishen Qubi Tongluo Decoction may reduce the level of inflammatory cytokines and improve joint inflammation by regulating miR-150-5p/SOCS1 pathway.
